响应面法在灵芝深层发酵产灵芝酸优化中的应用 被引量:1

摘要 目的:采用响应面法对影响灵芝酸产量的三种诱导物浓度进行优化,以提高深层发酵灵芝酸产量。方法:在单因素试验基础上,通过Design Expert软件设计CaCl 2、CuSO 4和褪黑素3种诱导物浓度,以灵芝酸含量与生物量之乘积为响应值优化灵芝深层发酵液诱导物加入浓度;菌丝体中灵芝酸含量测定采用香草醛-高氯酸法,菌丝体生物量测定采用称干重法。结果:当褪黑素为20μmol/L、CuSO 4为50μmol/L、CaCl 2为200μmol/L时,灵芝酸产量为所有试验组中的最高值,达到451.92 mg/L,比对照组增加了189%。响应面二次回归方程的预测值与验证试验结果基本一致。结论:通过响应面试验对灵芝酸诱导剂CaCl 2、CuSO 4和褪黑素浓度优化效果较好,诱导剂之间存在相互作用。 Objective:Response surface methodology was applied to optimize the concentration of three inducers in submerged fermentation broth in order to enhance the yield of ganoderic acid.Methods:Based on the single factor tests,response surface method was designed by Design Expert,the effect of the concentration of calcium chloride,copper sulfate and melationin on the the yield of ganoderic acid was studied.The mycelia was ground into powder in order to extract ganoderic acid with ethanol.The content of ganoderic acid was determined with vanillic aldehyde-perchloric acid and the yield of ganoderic acid was calculated on the basis of its content and mycelia biomass.Results:It showed that the production of ganoderic acids reached to the highest value of 451.92 mg/L in the concentration of calcium chloride concentration at 200μmol/L,copper sulfate concentration at 50μmol/L,melationin concentration at 20μmol/L.Under the condition of optimal combination,the yield of ganoderic acid was increased by 189%.Validation test results and the response surface fitting and forecast are in line with good income equation.Conclusion:It indicates that the experiment is effective,and there is interaction among inducers.
