摘要
通过轮状病毒(RV)灌胃感染SPF级7日龄BALB/c乳鼠,同时设立PBS阴性对照组和正常对照组,所有鼠均饲养在SPF级动物房,每日观察乳鼠腹泻情况。分别于接种后1、2、3、4、5、6、7、8、9、10d每组各处死5只乳鼠,在无菌环境中取乳鼠的小肠组织,通过qPCR方法检测病毒含量,HE染色观察并检测乳鼠小肠组织中RV载量及其组织病理变化,试剂盒检测乳糖酶活力。RV在感染第1天乳鼠的小肠组织中检测出病毒且相对含量表达较高;感染后前3d出现严重腹泻;HE染色可见乳鼠的小肠组织有损坏,小肠绒毛脱落,长度变短,PBS阴性对照组和正常对照组对照组对应结果均为阴性;乳糖酶活力检测较PBS阴性对照组和正常对照组降低。因此,建立RV感染BALB/c乳鼠继发性乳鼠乳糖不耐受动物模型,为研究乳糖不耐受症的发病机制及药物干预提供有力保障。
The 7-day-old SPF grade BALB/C suckling mice were gavaged with rotavirus(RV). At the same time, PBS negative control group and normal control group were set up, all mice were kept in SPF grade animal room to observe the diarrhea of suckling mice every day. On the 1 st, 2 nd, 3 rd, 4 th, 5 th, 6 th, 7 th, 8 th, 9 th and 10 th day after inoculation, respectively, 5 suckling mice were killed. The small intestine tissues of suckling mice were collected in sterile environment. The viral content was detected by qPCR method, RV load and the histopathological changes were observed by HE staining, and the activity of lactase was detected by commercial kit. RV was detected in the intestinal tissue of suckling mice on the first day of infection, and the relative content of RV was higher;severe diarrhea occurred in the first 3 d after infection;HE staining showed that the small intestine tissue of suckling mice was damaged, the villi of small intestine were shed off, and the length became shorter. The corresponding results of PBS negative control group and normal control group were negative;the lactase activity was lower than that of the PBS negative control group and normal control group. Therefore, the establishment of RV infected BALB/C suckling mice with secondary lactose intolerance animal model will provide a strong guarantee for studying the pathogenesis of lactose intolerance and drug intervention.
