马铃薯蛋白质提取工艺优化及分子量的测定 被引量:3

摘要 以马铃薯为原料,通过正交实验优化提取工艺,最终确定提取条件为料液比1∶2,碱溶性蛋白用pH=3.5提取,酸溶性蛋白用pH=8.5提取,总提取率为85.9%.提取出的蛋白质经过进一步纯化,利用凝胶层析法测定马铃薯蛋白质分子量,测量结果显示分离pH=8.5时,存在3种蛋白质,其分子量分别为1.2×10^4、7.8×10^4和4.0×10^4.分离pH=3.5时,存在1种蛋白质,其分子量为4.0×10^4. Taking potatoes as raw materials,the potato protein was extracted by alkali or acid isolation which was optimized by orthogonal experiment.Under the optimization extraction technology of alkali isolation pH 8.5,acid isolation pH 3.5 and 1∶2 to the ratio of raw and liquid,the sum recovery rate of protein was 85.9%.After further purified,the molecular weight of potato protein was determined by gel chromatography,the measurement results showed that three kinds protein were extracted by alkali isolation(pH 8.5),their molecular weight were 1.2×10^4 and 7.8×10^4 and 4.0×10^4;one kind of protein was extracted by acid isolation(pH 3.5),its molecular weight was 4.0×10^4.
