两种精子分析仪的检测性能评估 被引量:2

Evaluation of the detection performance of two sperm analyzers

摘要 目的探讨北昂(BEION S3)和瑞祺(CFT-9201)两种精子分析仪测试结果是否具有可比性,为检验结果互认提供依据。方法使用各自的质控品分别用两种精子分析仪重复分析10次,以比较质控品的有效性和仪器检测性能;分别用两种精子分析仪分析50例门诊患者的新鲜精液样本,并用配对t检验和Pearson或Spearman相关性分析比较两者的精子浓度、精子活动率、前向运动精子百分率(PR)及各项精子运动参数的差异及相关性。结果瑞祺精子分析仪和北昂精子分析仪分析瑞祺质控品[标称值为(22±3)×10^6/ml]的结果分别为(21.79±1.31)×10^6/ml和(24.41±2.90)×10^6/ml,两者有显著差异(P=0.03);而瑞祺精子分析仪和北昂精子分析仪分析北昂质控品[标称值为(52±2.07)×10^6/ml]的结果分别为(46.1±0.74)×10^6/ml和(45.34±2.53)×10^6/ml,两者没有显著差异,但均明显低于标称值。瑞祺精子分析仪和北昂精子分析仪分析50例精液样本的PR有显著差异,而精子浓度和活动率没有显著差异;精子运动参数除了直线速率(VSL)没有显著差异外,其余均有显著性差异;除直线性(LIN)和鞭打频率(BCF)没有显著相关性外,精液常规参数和其余精子运动参数均呈显著正相关。另外,北昂精子分析仪的几乎所有精液常规参数和精子运动参数的标准差和变异系数均高于瑞祺精子分析仪。结论不同精子分析仪厂家提供的质控品质量非常重要,应有标准化的机构认证为好;不同精子分析仪间存在系统误差,仪器参数设置需要标准化。而且,带有相差显微镜的精子分析仪性能要好于普通光学显微镜的精子分析仪。 Objective To investigate the comparability of the detection results of Beion S3 and Ruiqi CFT-9201 sperm analyzers,and then provide the evidence for the mutual recognition of the detection results.Methods Each of quality control products from Beion and Ruiqi corporations was analyzed for 10 times by two kinds of sperm analyzers,respectively,to compare the effectiveness of the quality control products and the performance of sperm analyzers.The fresh semen samples from 50 outpatients were detected by two kinds of sperm analyzers,respectively,and the differences and correlations of sperm concentration,motility,the percentage of progressive sperm(PR)and sperm motion parameters between the two sperm analyzers were analyzed with paired t test and Pearson or Spearman correlation analysis,respectively.Results The detection results of Ruiqi sperm analyzer and Beion sperm analyzer for the quality control product from Ruiqi Co.with nominal value of(22±3)×10^6/ml were(21.79±1.31)×10^6/ml and(24.41±2.90)×10^6/ml,respectively,and there was significant difference between them(P=0.03).While the detection results of Ruiqi sperm analyzer and Beion sperm analyzer for the quality control product from Beion Co.with nominal value of(52±2.07)×10^6/ml were(46.1±0.74)×10^6/ml and(45.34±2.53)×10^6/ml,respectively.There was no significant difference between them,but they were significantly lower than the nominal value.There was a significant difference in PR but not sperm concentration and motility of 50 semen samples detected by Ruiqi and Beion sperm analyzers.There were significant differences in sperm motion parameters except for straight-line velocity(VSL).There were significantly positive correlations of semen routine parameters and sperm motion parameters except for linearity(LIN)and beat-cross frequency(BCF)between the two sperm analyzers.In addition,the standard deviation and coefficient of variation of almost all semen routine parameters and sperm motion parameters of Beion sperm analyzer were higher than those of Ruiqi sperm analyzer.Conclusion The quality of quality control products provided by different sperm analyzer manufacturers is very important,and quality control products are better to be certified by the specified standardization organization.There are systematic errors between different sperm analyzers,indicating that the setting of instrument parameters needs to be standardized.Moreover,the performance of the sperm analyzer with a phase-contrast microscope is better than that with an ordinary optical microscope.
