摘要
肝细胞癌(hepatocellular carcinoma,HCC)占原发性肝癌的75%~85%,是一种起病隐匿、发展迅速、易复发、转移及耐药的恶性肿瘤。因此,探究HCC的复发、转移及有效的作用靶点是当前治疗的重中之重。肿瘤细胞进行代谢重编程使其在自身能量代谢、氨基酸代谢等方面具备了独特的特点。研究表明,尿素循环中的关键酶精氨酸代琥珀酸合成酶1(argininosuccinate synthase 1,ASS1)与HCC的侵袭和转移密切相关。本文结合该领域的最新研究进展探讨了ASS1的生化结构、功能调节及其在HCC进展中的作用和潜在价值,以期为HCC的精准治疗提供新的特异性分子治疗靶点,为新的治疗策略提供理论依据。
Hepatocellular carcinoma(HCC)accounts for 75%-85%of primary liver cancer,which is a malignant tumor with insidious onset,rapid development,easy recurrence,metastasis and drug resistance.Therefore,it is urgent to explore the recurrence,metastasis and effective targets of HCC.The metabolic reprogramming of tumor cells makes them have unique characteristics in energy metabolism and amino acid metabolism.Studies have shown that argininosuccinate synthase 1(ASS1),a key enzyme in the urea cycle,is closely related to the invasion and metastasis of HCC.Combine with the latest research progress in this field,we explored the biochemical structure and functional regulation of ASS1 and its role and potential value in the progression of HCC in this article,so as to provide theoretical evidences for new specific molecular therapeutic targets and new therapeutic strategies for accurate treatment of HCC.
