基于生物信息学分析癫痫相关基因在低级别胶质瘤患者预后中的作用 被引量:2

Effects of epilepsy-related genes on the prognosis of patients with low-grade gliomas based on bioinformatics analysis

摘要 目的基于生物信息学探讨癫痫相关基因对低级别胶质瘤(LCG)患者预后的影响,并初步挖掘其作用机制。方法从NCBI CEO数据库获取伴有或不伴有癫痫的LCG患者的芯片数据(CSE32534);用GEO2R算法比对筛选差异基因;利用DAVID数据库对癫痫相关基因进行CO功能和KECG通路分析;用STRINC数据库构建蛋白互作网络并多算法筛选Hub基因;利用GEIPA+TIMER数据库分析Hub基因对LCG患者预后及免疫细胞浸润的影响。结果共筛选出46个癫痫相关基因,主要富集在神经递质传递、关键启动子结合、胞外基质受体互作等过程;SYN1、GRIN2B、GRIA2、MAPT、SLC1A2、CRM5、RBFOX3、NRXN1是Hub基因,其上调可提高LCG患者存活率。结论多个癫痫相关基因可影响LCG患者预后,其机制可能与肿瘤内免疫细胞浸润调控有关。 Objective To investigate the effects of epilepsy-related genes on the prognosis of patients with low-grade gliomas(LCG)and to explore the active mechanism based on bioinformatics.Methods The microarray data-set GSE32534 containing LGG patients with or without epilepsy were obtained from NCBI-CEO database,and dif-ferential genes were acquired by CEO2R.Database DAVID was used to analysis the enrichment of epilepsy-relatedgenes in G0 module and KECG pathway.The interaction network of protein was constructed with database STRINGand multi algorithm was used to screen Hub genes.Databases CEIPA and TIMER were used to analyze the effects ofHub genes on LCG prognosis and infiltration of immune cells.Results A total of 46 epilepsy-related genes werescreened out which mainly enriched in the process of neurotransmitter secretion,core promoter sequence binding and EMC-receptor interaction.SYN1,GRIN2B,CRIA2,MAPT,SLC1A2,CRM5,RBFOX3,and NRXN1 were Hubgenes.Their up-regulation could improve the survival of LGG patients.Conclusions A variety of epilepsy-relatedgenes could affect the prognosis of LGG patients,and its mechanism may be closely associated with the regulation ofthe infiltration of immune cells in tumor.
