PRL对HC11细胞乳蛋白及其调节因子基因表达的影响 被引量:1

摘要 为了探讨PRL对大鼠乳腺上皮细胞中主要乳蛋白基因(CSN1S1、CSN2、LALBA)、GLUTs和细胞因子基因表达的影响,本试验以HC11细胞为载体,分别研究了催乳素对HC11细胞增殖、酪蛋白、葡萄糖转运蛋白和细胞因子基因表达的影响。结果表明:催乳素显著增加了HC11细胞αS1-CN,β-CN和α-LA基因的表达(P<0.05),但对GLUTs和炎症因子基因表达无影响(P>0.05),说明催乳素诱导的HC11细胞模型更适合于对乳蛋白合成和分泌有负面影响的因素及作用机制的研究,否则需要注意排除PRL干扰。 The purpose of this study was to investigate the effect of PRL on major casein genes(CSN1S1,CSN2,α-LA),GLUTS and cytokine genes expression in mammary epithelial cells.The HC11 cells were used as a carrier,the cell proliferation,content of casein and the genes expression of glucose transporters and inflammatory cytokines were studied.The results showed that the increased HC11αS1-CN,β-CN andα-LA gene expression were induced significantly by prolactin(P<0.05).But PRL has no influence on the GLUTs and inflammatory cytokines gene expression(P>0.05).It indicates that the prolactin-induced HC11 cell model is more suitable for the study of factors and mechanisms that have negative effects on milk protein synthesis and secretion.Otherwise,attention should be paid to exclude the interference of PRL.
