血浆LncRNA TUG1水平与冠心病患者冠状动脉病变程度相关性 被引量:13

The relationship between the level of plasma LncRNA TUG1 and the degree of coronary artery disease in patients with coronary heart disease

摘要 目的探讨长链非编码RNA (LncRNA)牛磺酸上调基因1(TUG1)在冠心病(CHD)患者血浆中的表达水平,分析其与冠状动脉病变程度的相关性。方法选取2017年9月-2020年1月上海市第六人民医院收治的96例CHD患者为CHD组,并选取同期本院96名体检健康者为对照组。采用实时荧光定量PCR(qRT-PCR)法检测所有受试者血浆LncRNA TUG 1表达水平;采用酶联免疫吸附法(ELISA)测定所有受试者血浆白细胞介素-6 (IL-6)水平;评估CHD患者Gensini评分;比较不同病变支数CHD患者血浆LncRNA TUG1、IL-6水平;Pearson法分析CHD患者血浆LncRNA TUG1水平与IL-6、Gensini评分的关系;采用受试者工作特征曲线(R0C)评价血浆LncRNA TUG1、IL-6水平对CHD的诊断价值。结果CHD组血浆LncRNA TUG1表达水平及IL-6水平均明显高于对照组,差异有统计学意义(t=12.030、12.271,P<0.05);CHD组Gensini评分为(10.68±3.06)分;随CHD患者冠状动脉病变支数增加,血浆LncRNA TUG1表达水平、IL-6水平均显著升高,差异有统计学意义(F=10.256、25.794,P<0.05);CHD患者血浆LncRNA TUG1水平与IL-6、Gensini评分均呈正相关(r=0.489、0.469,P<0.05);血浆LncRNA TUG1、IL-6诊断CHD的曲线下面积(AUC)分别为0.875、0.834,截断值分别为1.40、7.74pg/mL,相应灵敏度分别为79.2%、81.3%,特异度分别为84.4%、75.0%;两者联合诊断CHD的AUC为0.929,其灵敏度、特异度分别为88.5%、86.5%。结论LncRNA TUG1在CHD患者血浆中表达上调,可能在CHD发展中起重要作用,与Gensini评分、IL-6有关,LncRNA TUG1、IL-6有利于诊断CHD、评估患者疾病严重程度。 Objective To investigate the expression of long non-coding RNA(LncRNA) taurine up-regulated gene 1(TUG1) in the plasma of patients with coronary heart disease(CHD),and analyze its correlation with the degree of coronary artery disease.Methods A total of 96 CHD patients admitted to our hospital from September 2017 to January2020 were selected for the study,as CHD group,and 96 healthy persons as control group.The expression of plasma LncRNA TUG1 was detected by real-time fluorescent quantitative PCR(qRT-PCR);the level of interleukin-6(IL-6) in plasma was measured by enzyme-linked immunosorbent assay(ELISA);the Gensini score of CHD patients was evaluated;the levels of plasma LncRNA TUG1 and IL-6 in CHD patients with different pathological branches were compared;Pearson method was used to analyze the relationship between the level of plasma LncRNA TUG1 and the IL-6,Gensini score;in addition,the diagnostic value of plasma LncRNA TUG1 and IL-6 levels for CHD was evaluated by receiver operating characteristic curve(ROC).Results The expression level of LncRNA TUG1 and level of IL-6 in C.HD group were significantly higher than those in control group(t=12.030,12.271,P<0.05);the Gensini score of CHD group was(10.68±3.06).With the increase of the number of coronary artery lesions in CHD patients,the expression level of LncRNA TUG1 and level of IL-6 in plasma increased significantly(F=10.256,25.794,P<0.05).There was a positive correlation between the level of plasma LncRNA TUG1 and IL-6,Gensini score(r=0.489,0.469,P<0.05).The areas under curve(AUC) of plasma LncRNA TUG1 and IL-6 in the diagnosis of CHD were 0.875 and 0.834,respectively,and the cutoff values were1.40 pg/mL and 7.74 pg/mL,respectively.The corresponding sensitivities were 79.2% and 81.3%,respectively,and the specificities were 84.4% and 75.0%,respectively.In addition,the AUC of the combined diagnosis for CHD was 0.929,and the sensitivity and specificity were 88.5% and 86.5%,respectively.Conclusions The expression of LncRNA TUG1 was upregulated in the plasma of CHD patients,which might play an important role in the development of CHD,and it could be related to Gensini score and IL-6.LncRNA TUG1 and IL-6 could be helpful for the diagnosis of CHD and the evaluation of disease severity degree.
