沉默SETDBl对结直肠癌SW480细胞增殖、侵袭、迁移及凋亡的影响

Effects of SETDBl silencing on proliferation,invasion,migration and apoptosis of colorectal cancer SW480 cells

目的探讨沉默组蛋白赖氨酸甲基转移酶SETDBl对结直肠癌SW480细胞增殖、侵袭、迁移及凋亡的影响。方法qRT-PCR方法与Western blot方法检测人正常结肠黏膜上皮细胞NCM460与人结直肠癌细胞株SW480中SETDBl的表达。采用稳定转染方法筛选出沉默SETDBl基因的SW480细胞,克隆形成实验及流式细胞术检测沉默SETDBl对SW480细胞增殖与凋亡的影响;Transwell实验检测沉默SETDBl对SW480细胞侵袭迁移能力的影响。结果SETDBl在人结直肠癌细胞SW480中表达显著高于正常结肠黏膜上皮细胞NCM460。克隆形成实验及流式细胞术检测结果发现,沉默SETDBl基因能够促进SW480细胞凋亡,抑制增殖;Transwell实验检测结果发现,沉默SETDBl基因能够抑制SW480细胞侵袭和迁移。结论SETDBl在人结直肠癌细胞株SW480中高表达,沉默SETDBl基因能够抑制SW480细胞的增殖、迁移与侵袭,并促进凋亡。

Objective To investigate the effect of silencing histone lysine methyltransferase(SETDBl)on proliferation,invasion,migration and apoptosis of colorectal cancer SW480 cells.Methods qRT-PCR and Western blot were used to detect the expression of SETDBl in NCM460 and SW480 cells.SW480 cells silencing SETDBl gene were selected by stable transfection method.The effects of SETDBl silencing on the proliferation and apoptosis of SW480 cells were detected by colony formation assay and flow cytometry;the invasion and migration ability of SW480 cells were detected by Transwell assay.Results The expression of SETDBl in SW480 cells was significantly higher than that in NCM460 cells.The results of clone formation test and flow cytometry showed that silencing SETDBl gene could promote the apoptosis and inhibit the proliferation of SW480 cells;Transwell assay showed that the silencing of SETDBl gene could inhibit the invasion and migration of SW480 cells.Conclusion SETDBl is highly expressed in human colorectal cancer cell line SW480,and Silencing SETDBl gene can inhibit the proliferation,migration and invasion of SW480 cells,and promote apoptosis.
