Expression of B-cell lymphoma factor 3 gene mRNA in renal clear cell carcinoma and its relationship with prognosis

目的探讨B细胞淋巴瘤因子3(BCL3)基因mRNA在肾透明细胞癌中的表达及其与预后的关系。方法纳入癌症基因组图谱(TCGA)数据库截止2019年10月收录的肾脏正常组织(正常肾组织组,72例)及原发肾透明细胞癌组织(肾透明细胞癌组织组,533例),根据分期将原发肾透明细胞癌组织组分为Ⅰ期组(267例)、Ⅱ期组(57例)、Ⅲ期组(123例)、Ⅳ期组(84例)。再根据是否有淋巴结转移将256例组织分为无转移组(N0组,240例)和转移组(N1组,16例),根据基因表达信息将380例组织分为透明细胞A型(ccA)组(205例)及透明细胞B型(ccB)组(175例)。收集肾透明细胞癌患者的临床特征数据,包括肿瘤分期、透明细胞亚型、淋巴结转移状态及总生存(OS)期。同时收集正常肾组织组及肾透明细胞癌组织组中BCL3基因mRNA的表达水平并比较。采用Kaplan-Meier生存分析法对BCL3基因mRNA表达情况与肾透明细胞癌患者OS期的关系进行分析。结果BCL3基因mRNA在肾透明细胞癌组织组中的表达水平明显高于正常组织组(P=0.0004)。肾透明细胞癌组织Ⅰ~Ⅳ期组组织中BCL3基因mRNA表达水平均高于正常肾组织组(P<0.05);Ⅲ期、Ⅳ期组组织中BCL3基因mRNA的表达水平均高于Ⅰ期组(P<0.01)。N0组和N1组组织中BCL3基因mRNA表达水平比较差异无统计学意义(P=0.1115)。ccB组组织中BCL3基因mRNA表达水平高于ccA组(P=0.010)。Kaplan-Meier生存分析结果显示,BCL3基因mRNA的高表达预示肾透明细胞癌患者OS期更短(P<0.001)。结论BCL3基因mRNA在肾透明细胞癌组织中呈高表达,且这种高表达与更差的预后有关。

Objective To investigate the expression of B-cell lymphoma 3(BCL3)mRNA in renal clear cell carcinoma and its relationship with prognosis.Methods The normal kidney tissues(normal kidney tissue group,72 cases)and primary renal clear cell carcinoma tissues(renal clear cell carcinoma tissue group,533 cases)included in the TCGA database until October 2019 were selected.According to the relevant standards,primary renal clear cell carcinoma tissues group were divided into stageⅠgroup(267 cases),stageⅡgroup(57 cases),stageⅢgroup(123 cases),and stageⅣgroup(84 cases).According to whether there was lymph node metastasis,256 tissues were divided into non-metastasis group(N0 group,240 cases)and metastasis group(N1 group,16 cases).The 380 tissues were divided into clear cell type A(ccA)group(205 cases)and clear cell type B(ccB)group(175 cases)based on gene expression information.Clinical characteristic of renal clear cell carcinoma patients were collected,including tumor stage,clear cell subtype,lymph node metastasis status and overall survival(OS)time.At the same time,the expression levels of BCL3 gene mRNA in normal kidney tissues group and primar y renal clear cell carcinoma tissues group were collected and compared.Kaplan-Meier survival analysis method was used to analyze the relationship between BCL3 gene mRNA expression and OS time of patients with renal clear cell carcinoma.Results The expressions of BCL3 gene mRNA in the renal clear cell carcinoma tissue group was significantly higher than that in the normal tissue group(P=0.0004).The expressions of BCL3 gene mRNA in renal clear cell carcinoma tissues from stageⅠtoⅣgroup were higher than that in normal renal tissues(P<0.05).The expressions of BCL3 gene mRNA in the tissues of stageⅢandⅣgroup were higher than that in the stageⅠgroup(P<0.01).There was no statistically significant difference in the expressions of BCL3 gene mRNA between N0 group and N1 group(P=0.1115).The expressions of BCL3 gene mRNA in the ccB group were higher than those in the ccA group(P=0.010).Kaplan-Meier survival analysis results showed that the high expression of BCL3 gene mRNA indicated that patients with renal clear cell carcinoma had shorter OS time(P<0.001).Conclusion BCL3 gene mRNA is highly expressed in patients with renal clear cell carcinoma,which is associated with a worse prognosis.
