口服黄芪颗粒对老年痴呆大鼠基质金属蛋白酶-9和Occludin表达的影响 被引量:2

Effect of oral administration of Astragalus Granule on expression of matrix metalloproteinase-9 and Occludin in Alzheimer’s rats

摘要 目的研究老年痴呆大鼠口服黄芪颗粒后,其海马组织中的基质金属蛋白酶-9与紧密连接蛋白(Occludin)水平的变化。方法制备老年痴呆大鼠模型,将研究对象分为对照组、模型组、黄芪颗粒组与吡拉西坦组。黄芪颗粒组:口服黄芪颗粒1.4 g/kg·d,时间为28天,西药组:口服吡拉西坦0.43 g/kg·d,时间为28天,对照组与模型组同时口服相同剂量的生理盐水。用水迷宫评估大鼠的学习记忆能力的变化,免疫印迹测量海马组织中的基质金属蛋白酶-9与Occludin的含量。结果与对照组相比,模型组小鼠表现的行为能力明显减退,基质金属蛋白酶-9的含量(0.565±0.037)增加而Occludin的含量(0.181±0.019)减少(均P<0.05);口服黄芪颗粒后,其行为能力显著上升,基质金属蛋白酶-9的含量减少(0.341±0.018)而Occludin的含量(0.293±0.017)增加(均P<0.05)。黄芪颗粒组与吡拉西坦组相比差异无统计学意义(P>0.05)。结论黄芪颗粒能提高老年痴呆大鼠的学习记忆能力,其机制为降低基质金属蛋白酶-9的含量,增加Occludin的含量。 Objective To study the changes of matrix metalloproteinase-9 and Occludin in hippocampus of Alzheimer’s rats after the oral administration of Astragalus Granule.Methods Alzheimer’s rat model was established,and the experiment animals were divided into the control group,the model group,the Astragalus Granule group and the pyracetamide group.The rats in the Astragalus Granule group were given the oral administration of Astragalus Granule 1.4 g/kg·d for 28 days,and the rats in the pyracetamide group were given the oral administration of pyracetamide 0.43 g/kg·d for 28 days,while the rats in control group and the model group were given the oral administration of the same dose of normal saline.The changes of learning and memory ability were assessed by water maze,and the contents of matrix metalloproteinase-9 and Occludin in hippocampus were measured by Western blot.Results Compared with the control group,the learning and memory ability of the model group decreased,while the content of matrix metalloproteinase-9(0.565±0.037)increased and the content of Occludin(0.181±0.019)decreased(all P<0.05).After the oral administration of Astragalus Granule,the learning and memory ability increased significantly,the content of matrix metalloproteinase-9(0.341±0.018)decreased and the content of Occludin(0.293±0.017)increased(all P<0.05).There was no statistically significant difference between the Astragalus Granule group and the pyracetamide group(P>0.05).Conclusion Astragalus Granule can improve the learning and memory ability of Alzheimer’s rats.The mechanism is to reduce the content of matrix metalloproteinase-9 and increase the content of Occludin.
