子宫内膜VEGF在月经周期的表达变化研究 被引量:1

摘要 目的探讨子宫内膜血管内皮生长因子(VEGF)在月经周期的表达变化。方法选取以男方因素不孕为主的患者行子宫内膜活检或宫腔镜检查的同时留取的子宫内膜标本113个,分为月经期(20个)、增殖早(15个)、中(16个)、晚期(17个)、分泌早(15个)、中(14个)和晚期(16个)共7个组,采用免疫组化的方法对VEGF进行定位,观察染色情况,并用图像分析系统对子宫内膜切片进行分析,计算累积吸光度值,同时采用反转录聚合酶链反应(RT-PCR)法分析子宫内膜VEGF mRNA的表达。结果VEGF表达于子宫内膜的腺上皮细胞、间质细胞及血管内皮细胞。月经期子宫内膜VEGF表达较低,增殖期开始升高,增殖晚期达到最大,与其余各个时期比较差异有统计学意义(P<0.05)。分泌早期表达下降,较增殖晚期下降显著(P<0.05),但到分泌中期,VEGF表达再次升高,分泌中、晚期与增殖早、中期水平相似(P>0.05)。结论子宫内膜VEGF在整个月经周期都有表达,且呈现周期性变化:增殖晚期表达最强,月经期及分泌早期表达最低,增殖早、中期及分泌中、晚期皆呈较高水平。 Objective To explore the expression of the VEGF in human endometrium during the menstrual cycle.Methods About 113 endometrial specimens were obtained through endometrial biopsy or hysteroscopy.The expression of VEGF was analyzed by immunohistochemistry and the expression of VEGF mRNA was analyzed by RT-PCR in menstrual,proliferative and secretory phase.ResultsVEGF was expressed in cytoplasm in endometrial glandular epithelial cells,stromal cells and vascular endothelial cells.The expression in menstrual period was lower.Then it became stronger during proliferative phase.Late proliferative phase had the highest expression and the difference with the other menstrual period was significantly(P<0.05).Early secretory phase expression began to decline significantly compared with the late proliferation phase(P<0.05).Mid and late secretory phase became stronger,but it was still similar to the early and mid proliferative phase(P>0.05).Conclusion Endometrial VEGF was expressed throughout the menstrual cycle,and presented periodic changes:the expression was strongest in the late proliferative phase and was lowest in the menstruation and early secretory phase.
