免疫荧光成像技术在骨架细胞生物学实验教学中的应用

摘要 将间接免疫荧光技术应用到细胞骨架的观察实验教学中,以48孔板培养细胞,控制细胞密度为50%~80%,以鼠抗人tubulin的单克隆抗体和FITC标记的羊抗鼠的荧光二抗对微管蛋白进行间接免疫标记,通过可视倒置荧光显微镜观察细胞骨架的形态和分布,提高了教学效果,增强了学生的科研兴趣和综合素质。 Cytoskeleton observation of is one of the classical teaching items for cell biology experiment of undergraduates,which is usually carried out by comassie brilliant blue staining of onion inner epidermal cells.In order to improve the experimental effect,immune fluorescent technology had been applied accordingly.The primary antibody was anti-βtubulin IgG,and the secondary antibody was goat anti-mouse FITC.Cells were cultured in 48 wells plate to a 50%~80%culture density.The distribution and morphology of cytoskeleton could be observed by inverted fluorescence microscope.This application was not only led to an improving result,but also stirred up the scientific research interest and comprehensive quality of students,which encouraged their creative thinking.
