摘要
为建立黑加仑提取物中飞燕草素葡萄糖苷、飞燕草素芸香苷、矢车菊素葡萄糖苷、矢车菊素芸香苷等4种主要花色苷成分的高效液相色谱法(HPLC)检测方法,采用Inertsil ODS-3色谱柱(4.6 mm×250.0 mm,5μm),流动相为2%磷酸水溶液∶色谱乙腈(310∶40,体积比),等度洗脱,流速1.0 mL/min,检测波长540 nm,柱温30℃。结果表明,飞燕草素-3-葡萄糖苷、飞燕草素-3-芸香苷、矢车菊素-3-葡萄糖苷、矢车菊素-3-芸香苷分别在5.56~278.00、5.64~282.00、5.78~289.00、5.80~290.00μg/mL范围内线性关系良好,R2≥0.9995。加样回收率为98.69%~100.89%,RSD均在1.5%以内。该方法简便准确,可以用于黑加仑提取物中主要花色苷的定量分析。
In order to establish an HPLC method for the determination of delphinidin-3-O-glucoside,delphinidin-3-O-rutinoside,cyanidin-3-O-glucoside,cyanidin-3-O-rutinoside of blackcurrant extract,Inertsil ODS-3 column(4.6 mm×250.0 mm,5μm)was used,with 2%phosphoric acid solution-chromatographic acetonitrile(310∶40,volume ratio)as the mobile phase for isocratic elution.The flow rate was 1.0 mL/min.The detection wavelength was at 540 nm.The column temperature was 30℃.The results showed that delphinidin-3-O-glucoside,delphinidin-3-O-rutinoside,cyanidin-3-O-glucoside,cyanidin-3-O-rutinoside had a good linear relationship respectively in the range of 5.56-278.00μg/mL,5.64-282.0μg/mL,5.78-289.00μg/mL,5.80-290.00μg/mL with R2≥0.9995.The average recovery was 98.69%-100.89%with RSD less than 1.5%.This method is simple,reliable and can be used for the quantitative analysis of the main anthocyanins in the blackcurrant extract.
