摘要
为了探索牛病毒性腹泻病毒(Bovine viral diarrhea-mucosal virus,BVDV)NS2-3基因的生物学特征,试验以不同生物型(ncp型和cp型)BVDV NS2-3基因序列及其蛋白氨基酸序列为研究对象,应用生物信息学软件对不同生物型BVDV的NS2-3基因序列同源性、遗传进化关系、磷酸化位点、糖基化位点、酰基化位点、信号肽、亚细胞定位、同源重组及B细胞线性表位进行预测分析。结果表明:ncp JL株NS2-3基因核苷酸序列与ncp PI285株同源性较高,cp Singer Arg株NS2-3基因核苷酸序列与cp/ncp NADL株同源性较高;ncp JL株与ncp Nebraska株及ncp PI285株遗传进化关系较近,cp Singer Arg株与cp/ncp NADL株、cp BVDV-1株遗传进化关系较近;不同生物型NS2-3蛋白所含磷酸化位点及糖基化位点数量不同,但均含有信号肽;ncp JL株NS2-3蛋白定位于宿主细胞核,而cp Singer Arg株NS2-3蛋白定位于宿主细胞核和宿主细胞质中;ncp JL株和cp Singer Arg株都与cp/ncp NADL标准株发生了基因重组;不同生物型BVDV都含有其特异线性表位。说明不同生物型BVDV的NS2-3蛋白在结构及理化性质等方面具有差异,可作为区分ncp型及cp型BVDV的指标。
In order to explore the biological characteristics of BVDV NS2-3 gene, the gene sequences and protein amino acid sequence of different biological type(NCP and cp) BVDV NS2-3 were selected as the research object in this experiment, the sequence similarity, homologous evolutionary relationship,phosphorylation site,glycosylation site,acylation site,signal peptide,subcellular localization and B cell linear epitopes of NS2-3 gene were predicted and analyzed by bioinformatics software. The results showed that the nucleotide sequence of NS2-3 gene of ncp-type BVDV-JL had higher homology with the ncp-type BVDV-P1285, and the nucleotide sequence of NS2-3 gene of cp-type BVDV-Singer Arg had higher homology with cp-type/ncp-type BVDV-NADL. The genetic evolution of ncp-type BVDV-JL and ncp-type Nebraska/ncp-type PI285 was relatively close. The genetic evolution of cp-type BVDV-Singer Arg and cp-type/ncp-type BVDV-NADL strain or cp-type BVDV-1 strain was relatively close. The number of phosphorylation sites and glycosylation sites in NS2-3 proteins of different biological types was different, but they all contained signal peptides. The NS2-3 protein of ncp type BVDV-JL was localized in the host cell nucleus, while the NS2-3 protein of cp type BVDV-Singer Arg strain was localized in the host cell nucleus and host cytoplasm. Both ncp-type BVDV-JL and cp-type BVDV-Singer Arg were genetically recombined with cp/ncp-type BVDV-NADL standard strain. Different biotypes of BVDV contained specific linear epitopes. These results indicated that the NS2-3 protein of different biological types of BVDV was different in structure and physicochemical properties, which could be used as an indicator to distinguish NCP type and CP type BVDV.
