UFLC-MS/MS法测定句容蝉花中核苷类成分肌苷、尿苷的含量

摘要 目的:建立同时测定蝉花中肌苷、尿苷含量的方法。方法:采用Shim-pack XP-ODSⅢ(1.6μm,75 mm×2.0 mm)为色谱柱,甲醇-水(5∶95)流动相,流速:0.2 mL·min^-1,柱温:40℃,进样量:1μL,多反应监测(MRM)和电喷雾离子化(ESI+)。结果:肌苷和尿苷分别在2.04~13.60 ng·mL^-1、15.72~104.80 ng·mL^-1范围内线性关系良好(R 2>0.99)。肌苷和尿苷的平均回收率分别为90.58%、91.72%。结论:该方法具有测定快速、选择性好、灵敏高的特点,可用于蝉花的质量控制。 Objective To o establish an UFLC-MS/MS method for the simultaneous determination of inosine and uridine in Jurong Cordyceps cicadae.Methods Shim-pack XP-ODSⅢC18 column(2.0 mmL.D×75 mm,1.6μm),the methanol and water(5∶95)as mobile phase.The flow rate was 0.2 mL·min^-1 and the column temperature was 40℃.MS instrument was equipped with positive electrospry ionization mode(ESI+)and multiple reaction monitoring(MRM).Result Inosine and uridine had good linear relationship in the range of 2.04~13.60 ng·mL^-1、15.72~104.80 ng·mL^-1(R 2>0.99).The average recovery rate of Inosine and uridine was 90.58%and 91.72%.Conclusion This method has the characteristics of fast selectivity and high sensitivity,and can be used for quality control of Cordyceps cicadae.
