枸杞子对光损伤小鼠视网膜的保护作用 被引量:5

摘要 以光损伤小鼠为模型,探讨不同剂量的枸杞多糖(lycium barbarum polysaccharide,LBP)粗提物(280、370、460 mg/kg)对光损伤小鼠视网膜形态、功能和抗氧化能力的影响。对小鼠连续干预8周后造模,进行视网膜电图(electroretinogram,ERG)和光学相干断层成像(optical coherence tomography,OCT)相关视网膜生理指标检测。ERG结果显示,通过比较0.01 b波和3.0 b波振幅,药物干预组小鼠视网膜对光的敏感程度要高于模型组。OCT检测结果表明,枸杞多糖粗提物各剂量组均较模型组视网膜色素上皮(retinal pigment epithelium,RPE)层细胞平滑,说明枸杞多糖粗提物对小鼠光损伤视网膜的形态损伤具有保护作用。视网膜抗氧化指标测定结果显示,枸杞多糖粗提物高剂量组的抗氧化效果好于低剂量组,呈剂量依赖性。研究结果显示,枸杞多糖粗提物对光损伤小鼠视网膜具有较好的保护作用。 This study used light damaged mice as models,and discussed the protective effect on the retina of lycium barbarum polysaccharide(LBP)crude extract from Fructus lycii,whose concentrations were 280,370 mg/kg and 460 mg/kg.The influences were discussed of LBP crude extract on the structure,function and antioxidant capacity of the retina.After mice were constantly administered with LBP crude extract for 8 weeks,the model was built and the pertinent retina physiological index tests were conducted,such as electroretinogram(ERG)and optical coherence tomography(OCT).In addition,the results of ERG showed that,comparing with the amplitude of 0.01 b wave and 3.0 b wave,the light sensitivity of the drug intervention group was higher than the model group.The results of OCT showed that retinal pigment epithelium(RPE)layer of each dose groups was smoother than that of model group,which illustrated that LBP crude extract had a protective effect on morphological damage of the light-damaged mice.The results of anti-oxidant index of retina showed that the antioxidant effect of the high-dose group of LBP crude extract was better than that of the low-dose group.The research results showed that LBP crude extract had a better protective effect on retina of light damaged mice.
