APOC1蛋白在胃癌中的表达及临床意义

Expression of APOC1 in gastric cancer and its clinical significance

目的探讨载脂蛋白C1(APOC1)在胃癌中的表达水平及其与临床病理特征的关系。方法应用TCGA数据库分析胃癌组织中APOC1的表达情况。收集70例胃癌组织和癌旁正常胃黏膜组织标本,采用免疫组织化学法和Western blot检测两组标本中APOC1蛋白的表达情况,分析APOC1蛋白表达的阳性表达率与胃癌患者的性别、年龄、分化程度、肿瘤直径、Lauren’s分型、淋巴结转移、TNM分期等临床病理特征的关系;采用Kaplan-Meier Plotter对胃癌患者的生存情况进行分析。结果通过TCGA数据库分析发现APOC1在胃癌中的表达高于癌旁正常胃黏膜组织。免疫组化结果显示APOC1蛋白在胃癌组织中的表达显著高于癌旁正常胃黏膜组织;Western blot实验显示在新鲜胃癌组织中检测到APOC1蛋白表达水平高于癌旁组织。APOC1蛋白在胃癌中的表达与TNM分期、淋巴结转移呈明显正相关;与患者年龄、性别、肿瘤直径、Lauren’s分型和分化程度无关;生存分析发现APOC1低表达组的OS、FP、PPS均显著长于高表达组(P<0.05)。结论 APOC1在胃癌组织中表达上调且与胃癌进展相关,其异常表达与胃癌患者的预后不良密切相关。APOC1有望成为胃癌诊断和评估预后的有效指标。

Objective To investigate the expression level of apolipoprotein C1( APOC1) in gastric cancer and its relationship with clinicopathological characteristics. Methods The expression of APOC1 in gastric cancer tissues was analyzed by TCGA database. The expression of APOC1 protein in specimens of 70 gastric cancer tissues and adjacent normal gastric mucosa was detected by immunohistochemistry and Western blot. Relationship between the positive expression rate of APOC1 protein and gender,age,differentiation degree,tumor diameter,Lauren’s classification,lymph node metastasis,TNM staging and other clinicopathological features were analyzed;Kaplan-Meier analysis was used to determine the survival of gastric cancer patients. Results The expression of APOC1 in gastric cancer was higher than that in normal gastric mucosa. The results of immunohistochemistry showed that the expression of APOC1 protein in gastric cancer tissues was significantly higher than that in adjacent normal gastric mucosa tissues. Western blot analysis showed that the level of APOC1 protein expression in fresh gastric cancer tissues was higher than that in the adjacent tissues. The expression of APOC1 protein in gastric cancer was positively correlated with TNM stage and lymph node metastasis;it was not related to patients’ age,gender,tumor diameter,Lauren’s typing and differentiation;OS,FP and PPS in APOC1 low expression group were significantly longer than those in high expression group. Conclusion The expression of APOC1 is up-regulated in gastric cancer and that is associated with the progression of gastric cancer. The abnormal expression of the APOC1 is closely related to the poor prognosis of the patients. APOC1 is expected to be an effective indicator for the diagnosis and prognosis evaluation of gastric cancer.
