摘要
目的:根据板蓝根和大青叶中有效成分靛玉红的性质,选择不同的溶媒和提取方法,优化其提取工艺的最佳条件。方法:分别用水和乙醇为溶媒,用正交试验设计法,采取乙醇回流、水提醇沉等提取方法,以靛玉红含量为指标,考察不同的实验方法和条件对靛玉红的影响,优选最佳的提取工艺条件。结果:水煎煮提取的最佳条件:D3 B2 A3 C1,即加8倍量的水提取2次,每次2 h,提取温度为100℃。乙醇回流提取的最佳条件是A3 B2 D2 C2,即加7倍量80%乙醇回流提取2次,每次80 min。结论:乙醇回流法提取板蓝根和大青叶中有效成分靛玉红是水煎煮法提取靛玉红的97倍。
Objective:According to the nature of the active ingredient indirubin in Isatidis Radix and Isadidis Folium,we select different solvents and extraction methods to optimize the optimal conditions of the extraction process.Methods:Considering the effects of different experimental methods and conditions on indirubin by the indirubin content as the detective marker,the optimum are chosen by orthogonal test,using the water and ethanol as solvents,ethanol reflux extraction,ethanol subsiding method and etc as extraction method.Results:The best conditions for water decoction extraction process is D3 B2 A3 C1.That is,8 times the amount(V/W)of water was added to the medicinal material and extracted for 2 times,two hours each time,the extraction temperature was 100℃.The best conditions for ethanol reflux extraction process is:A3 B2 D2 C2.That is,7 times the amount(V/W)of 80%ethanol solution was added to the medicinal material and extracted twice by reflux,80 minutes each time.Conclusion:The effective component of the extract of Radix Isatidis and Daqingye by ethanol reflux method is 97 times of the extraction of indirubin red by water decoction.
