循环miRNA与寻常型银屑病不同中医证候的相关性研究

Correlation Between Circulating miRNA and Different Tradition Chinese Medicine Syndromes of Psoriasis Vulgaris

目的探讨血浆miR-146a、miR-155、miR-326在寻常型银屑病及其不同中医证型中的表达水平以及与中医辨证分型的关系。方法采用实时荧光定量聚合酶链反应(PCR)法检测分别检测60例寻常型银屑病患者(血热证30例、血瘀证15例、血燥证15例)血浆中miRNA,并与健康对照组对比分析及组间比较。结果血浆miR-146a、miR-155在寻常型银屑病患者中的表达水平升高,且与疾病的严重程度呈正相关;与血瘀证和血燥证患者相比,血热证患者血浆miR-146a、miR-155相对表达量显著升高,差异有统计学意义。血浆miR-326在寻常型银屑病患者中的相对表达量与健康对照相比差异无统计学意义,但血燥证银屑病患者血浆miR-326的相对表达量较血热证和血瘀证明显减少,不同证型间比较差异有统计学意义。结论循环血浆miRNA在不同证型寻常型银屑病患者中存在表达差异,可作为寻常型银屑病中医辨证分型的参考依据。

Objective In order to investigate the expression levels of plasma miRNA-146a,miRNA-155 and miRNA-326 in psoriasis vulgaris and its different traditional Chinese medicine(TCM)syndromes and their relationships TCM syndrome differentiation.Methods Sixty cases with psoriasis vulgaris(30 cases with blood heat syndrome,15 cases with blood stasis syndrome and 15 cases with blood dryness syndrome)were detected by real-time fluorescence quantitative polymerase chain reaction(RT-qPCR).Results The expression levels of plasma miR-146a and miR-155 in patients with psoriasis vulgaris were increased,which was positively correlated with the severity of the disease.Compared with patients with blood stasis syndrome and blood dryness syndrome,the relative expression levels of plasma miR-146a and miR-155 in patients with blood heat syndrome were significantly increased,which was statistically significant.The expression of plasma miR-326 in patients with psoriasis vulgaris was decreased,which was statistically significant,and the relative expression level of plasma miR-326 in patients with psoriasis with blood dryness syndrome was lower than that in patients with blood heat syndrome and blood stasis syndrome,and the comparison between different syndromes was statistically significant.Conclusion There are differences in the expression of circulating plasma miRNA in patients with different syndromes of psoriasis vulgaris,which can be used as references for TCM differentiation of psoriasis vulgaris.
