辛开苦降法对脾胃湿热证小鼠炎症因子和免疫功能的影响 被引量:4

Effect of Xinkai Kujiang Method on Inflammatory Factors and Immune Function of Mice with Spleen-Stomach Damp-heat

摘要 目的观察连朴饮对小鼠脾胃湿热证的炎症因子与凋亡基因、抑癌基因蛋白表达的调节作用,探讨辛开苦降法对脾胃湿热证免疫功能的影响。方法采用内因湿热法即高脂高糖饮食复制脾胃湿热证动物模型。SPF级昆明小鼠,雌雄各半随机分成6组,即正常组,模型组,黄芩滑石汤组,连朴饮低、中、高剂量组(6.2、12.4、24.8 g/kg),每组10只。用ELISA法检测干扰素-γ(interferen-gamma,IFN-γ)、白细胞介素4(IL-4)的含量;用HE染色法观察胃肠组织形态学的变化;用蛋白质印迹法检测Bcl-2、p53的蛋白表达水平。结果与正常组相比,模型组小鼠血清中IFN-γ、IL-4、IFN-γ/IL-4值明显升高(P<0.05)。与模型组比较,连朴饮各剂量组均显著降低小鼠血清中上述指标的含量(P<0.05)。高剂量组的指标降低最明显,其次是中剂量组,低剂量组的指标降低的较少。结论连朴饮能降低IFN-γ、IL-4值,抑制Bcl-2、p53蛋白的表达,其发挥作用的机制可能是降低机体的炎症反应,改善免疫状态,恢复免疫平衡。 Objective To observe the regulating effect of Lianpu Yin on the expression of inflammatory factors,apoptosis genes and tumor suppressor genes in the spleen-gastric damp-heat syndrome of mice,and to explore the effect of Xinkai Kujiang method on the immune function of spleen-gastric damp-heat syndrome.Methods Reproduction of animal model of spleen and stomach,dampness and heat syndrome by using internal cause as heat method,that is,high fat and high sugar diet.SPF Kunming mice were divided into 6 groups semi-randomly,male and female,namely normal group,model group,Huangqin Huashitang group,Lianpu Yin high,medium and low dose groups(6.2,12.4,24.8 g/kg),each group 10 only.The content of interferon-γ(interferen-gamma,IFN-γ)and interleukin 4(IL-4)were detected by ELISA;the morphology of gastrointestinal tissue was observed by HE staining;Detection of Bcl-2 and p53 protein expression levels by Western Blot method.Results Compared with the normal group,the serum levels of IFN-γ,IL-4,IFN-γ/IL-4 in the model group were significantly increased(P<0.05).Compared with the model group,each dose group of Lianpu Yin significantly reduced the content of the above indexes in mouse serum(P<0.05).The index of the high-dose group decreased most obviously,followed by the medium-dose group,and the index of the low-dose group decreased less.Conclusion Lianpu Yin can reduce the value of IFN-γand IL-4,inhibit the expression of Bcl-2 and p53 protein,and its mechanism may be to reduce the body’s inflammatory response,improve the immune state,and restore the immune balance.
