脾瘅宁对糖调节受损大鼠脂肪中NF-κB通路调控作用的实验研究 被引量:2

摘要 目的阐述脾瘅宁通过抑制TNF-α/NF-κB的过度活化改善葡萄糖调节受损大鼠脂、糖代谢,探讨其抗炎机制。方法用高糖高脂饲料喂养的方法,诱导葡萄糖调节受损大鼠模型,并给予脾瘅宁水煎剂经口灌胃干预,测定给药前后血糖、体质量等观察指标;采用RT-PCR和western-blot方法对TNF-α/NF-κB mRNA及蛋白表达水平进行检测。结果与空白对照组比较,模型对照组、脾瘅宁组和二甲双胍对照组大鼠血糖、体质量及脂肪组织中TNF-α、NF-κB mRNA及蛋白的表达水平均显著升高,差异有统计学意义(P<0.01);与模型对照组比较,脾瘅宁组和二甲双胍对照组大鼠血糖、体质量及脂肪组织中TNF-α、NF-κB mRNA及蛋白的表达水平均显著降低,差异有统计学意义(P<0.05或0.01);除空腹血糖外,脾瘅宁组和二甲双胍对照组大鼠各指标间无统计学差异。结论脾瘅宁抑制TNF-α/NF-κB信号通路的过度活化作用可能是其抑制慢性炎症反应,调节脂代谢和糖代谢的作用机制之一。 Objective To investigate the effect of Pidan Ning on lipid and glucose metabolism in impaired rats by inhibiting the excessive activation of TNF-α/NF-κB,and to explore its anti-inflammatory mechanism.Methods High glucose and high fat diet was used to induce the impaired rat model of glucose regulation.The blood glucose and body weight were measured before and after the administration.The expression of TNF-α/NF-κB mRNA and its protein were detected by RT-PCR and western-blot.Results Compared with the blank control group,the levels of TNF-α,NF-κB mRNA and protein in the model control group,the Pidan Ning group and metformin control group were significantly higher than those in the control group,and the expression levels of TNF-α,NF-κB mRNA and protein in adipose tissue were significantly increased.The difference was statistically significant(P<0.01).Compared with the model control group,the blood glucose,body weight and the expression of TNF-α,NF-κB mRNA and protein in the adipose tissue of the rats in the Pidan Ning group and metformin control group were significantly decreased(P<0.05 or P<0.01).Except for fasting blood glucose,there was no statistical difference between Pidan Ning group and metformin control group.Conclusion The inhibition of excessive activation of TNF-α/NF-κB signaling pathway of Pidan Ning may be one of its mechanisms for inhibiting chronic inflammation and regulating lipid metabolism and glucose metabolism.
