Expressions and clinical significance of Twist and E-cadherin in human osteosarcoma tissues

摘要 目的通过测定Twist及E-cadherin分别在骨肉瘤、骨软骨瘤组织中的表达情况,试评价两者在骨肉瘤和骨软骨瘤发生、发展过程中的临床意义及其相关性。方法用免疫组织化学SP法检测50例骨肉瘤组织和12例骨软骨瘤组织中Twist、E-cadherin的表达情况,采用SPSS 23.0对其结果作统计分析,探讨其意义。结果(1)50例骨肉瘤中41例Twist表达阳性,阳性率82.00%;12例骨软骨瘤中仅2例Twist为阳性染色,阳性率16.67%。两组间比较差异有统计学意义(P<0.01)。Twist在骨肉瘤不同病理分型中表达差异无统计学意义(P>0.05),而在不同病理分级中表达差异有统计学意义(P<0.05)。(2)50例骨肉瘤中有17例E-cadherin为阳性,阳性率为34.00%;12例骨软骨瘤中有9例E-cadherin表达阳性,阳性率为75.00%。两组间比较差异有统计学意义(P<0.05)。E-cadherin在骨肉瘤不同病理分型和分级中表达差异无统计学意义(P>0.05)。(3)转移组骨肉瘤Twist阳性表达率为93.10%(27/29),非转移组阳性率为46.15%(6/13),两组阳性率比较差异有统计学意义(P<0.05)。转移组骨肉瘤E-cadherin阳性表达率为24.14%(7/29),非转移组阳性率为61.54%(8/13),两组E-cadherin表达的阳性率比较差异有统计学意义(P<0.05)。Twist与E-cadherin在骨肉瘤中的表达呈明显负相关(rs=-0.457,P=0.001<0.01)。结论骨肉瘤恶性程度越高Twist表达越高;Twist和E-cadherin可作为临床上辅助判断骨肉瘤预后的指标;Twist可能通过抑制E-cadherin表达促进骨肉瘤侵袭转移。 Objective To study the expressions and correlations of Twist and E-cadherin respectively in osteosarcoma and osteochondroma,and to evaluate their clinical significance in the occurrence and progression of osteosarcoma and osteochondroma.Methods Expressions of Twist and E-cadherin in 50 cases of osteosarcoma and 12 cases of osteochondroma were detected using immunohistochemistry method.Expression differences and clinical significance were analyzed by SPSS 23.0.Results(1)Positive Twist expressions were noted in 41 among 50 osteosarcoma patients with a positive immunostaining rate of 82.00%,while in 2 among 12 osteochondroma patients with a positive immunostaining rate of 16.67%;significant differences were observed between the two groups(P<0.01).No obvious differences in different pathological classifications of the osteosarcoma were seen in the Twist expressions(P>0.05),but not in different pathological gradings(P<0.05).(2)Positive E-cadherin expressions were noted in 17 among 50 osteosarcoma patients with a positive immunostaining rate of 34.00%,while in 9 among12 osteochondroma patients with a positive immunostaining rate of 75.00%;significant differences were observed between the two groups(P<0.05).No obvious differences in different pathological classifications or gradings of the osteosarcoma were seen in the E-cadherin(P>0.05).(3)The positive Twist immunostaining rate of the osteosarcoma transfer group was 93.10%(27/29),and 46.15%(6/13)in the non-transfer group;significant differences were observed between the two groups(P<0.05).The positive E-cadherin immunostaining rate of the osteosarcoma transfer group was 24.14%(7/29),and 61.54%(8/13)in the non-transfer group;significant differences were observed between the two groups(P<0.05).Twist expressions were negatively correlated with E-cadherin expressions in osteosarcoma(rs=-0.457,P=0.001<0.01).Conclusions Higher osteosarcoma malignancy brings higher Twist expressions.Twist and E-cadherin can be used as prognostic indicators for osteosarcoma.Twist may promote the invasion and metastasis of osteosarcoma by inhibiting E-cadherin expressions.
