三明治型电化学适体传感器的构建以及酶信号放大检测凝血酶

摘要 基于合成的纳米材料和酶信号放大技术构建了对凝血酶检测的电化学适配子传感器。利用纳米碳球电沉积纳米金修饰的核酸适配子作为传感平台以及二氧化硅纳米球标记的核酸适配子和信号探针与凝血酶特异性结合形成了三明治型的适配子传感器,通过亲和素标记的辣根过氧化物酶(HRP)催化对苯二酚和过氧化氢(HQ+H2O2)检测体系产生的电化学信号,实现对凝血酶的检测。该传感器在0.0001~100 ng/mL的凝血酶浓度范围内呈现良好的线性关系,其线性回归方程为I(μA)=-39.63-6.52lgc(ng/mL),相关系数R^2=0.994,检出限为0.084 pg/mL,且溶菌酶等无明显干扰。该传感器可用于凝血酶的检测。 An electrochemical aptasensor for detection of thrombin was constructed based on nanomaterials and enzyme signal amplification technique.The sandwich-type was established by carbon nanospheres and electrodeposited gold nanoparticles modified aptamer as the sensing platform,silicon dioxide nanoparticles modified aptamer and signal probe specifically combined with thrombin.Thrombin was detected by utilizing the electrochemical response from biotin labeled HRP catalyzed HQ+H2O2 detection system.The aptasensor showed a good linear relationship between current and thrombin concentration in the range of 0.0001~100 ng/m L with a limit of detection of 0.084 pg/m L.The linear regression equation was I(μA)=-39.63-6.52 lgc(ng/m L),and the coefficient R^2 was 0.994.Lysozyme had no obvious interference.This strategy can be used for detection of thrombin.
