摘要
目的:观察长期笑气(N2O)吸入对大鼠肝脏毒副作用的影响。方法:将10只Wistar雄性大鼠随机分为笑气暴露组和吸氧对照组,笑气暴露组暴露在N2O和O2(1:1)混合气体中,吸氧对照组暴露在氧气中,氧浓度控制在50%。2组气流速度均为1.5L/min,吸入时间120min/d,连续60d。全自动生化分析仪检测大鼠血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)水平;TUNEL法检测大鼠肝细胞凋亡;HE染色观察大鼠肝脏组织学特点改变。结果:笑气暴露组大鼠ALT、AST均高于吸氧对照组,其中ALT检测值2组间差异有统计学意义(P<0.05)。笑气暴露组大鼠肝细胞凋亡与吸氧对照组间差异无统计学意义(P=0.408)。笑气暴露组大鼠肝脏组织学特点与吸氧对照组相比无明显改变。结论:长期笑气吸入可导致大鼠肝功能损伤,或可引起肝细胞凋亡趋势增加,但肝脏组织学特点无明显改变。
Objective:To investigate the toxicity and side effects of long-term nitrous oxide inhalation on the livers of rats.Methods:The Wistar rats were divided into two groups randomly.The experimental group was exposed to 1.5L/min N2O with 1:1 O2 and the control group was exposed to O2(50%concentration)for 120min per day,altogether for 2 months.The analanine transaminase(ALT)and aspartate aminotransferase(AST)of the rats were tested by automatic biochemical analyzer.The TUNEL staining was used to detect the hepatocyte apoptosis of rats.The histology of the livers was evaluated by hematoxylin and eosin(HE)staining.Results:Comparing with the control group,the level of ALT of the experimental group was increased significantly(P<0.05).While there was no statistically significant between the two groups in the level of AST and the trend of hepatocyte apoptosis(P=0.309,0.408).Conclusion:Long-term inhalation of N2O could lead to the damage of liver function and may increase the tendency of hepatocyte apoptosis of the rats,but the histological characteristics of their liver tissues did not change significantly.
