大熊猫主食竹、粪便及血清中的短链脂肪酸的测定及分析

摘要 通过气相色谱—质谱联用(GC/MS)定量分析苦竹、大熊猫粪便及血清中短链脂肪酸(SCFAs)含量。对样品进行衍生化预处理、萃取,并进行GC/MS快速分析。样品中的12种SCFAs,包括甲酸、乙酸、丙酸、异丁酸、丁酸、异戊酸、2-甲基丁酸、戊酸、2-甲基戊酸、4-甲基戊酸、正己酸、正庚酸得到鉴定并准确定量。结果显示,苦竹中的短链脂肪酸在其不同部位存在明显差异,其主要短链脂肪酸与粪便中的基本一致,均以乙酸、甲酸和丙酸为主;血清中的短链脂肪酸以甲酸、乙酸、丙酸为主。在此基础上,探讨了苦竹中短链脂肪酸对大熊猫肠道和机体发育所产生的生理作用,能维持不同年龄个体肠道功能的平衡性和完整性,以及提高雌性泌乳期的产奶量和体重。本研究利用GC/MS测定了大熊猫可食竹苦竹、大熊猫粪便及血清中短链脂肪酸的含量,可为大熊猫食性及生理等研究提供数据支撑。 The content of short chain fatty acids(SCFAs)in Pleioblastus amarus,and feces and serum of giant panda were measured using gas chromatography-mass spectrometry(GC/MS).The samples were derivatized,extracted and quantified.Twelve SCFAs,viz.formic acid,acetic acid,propionic acid,isobutyric acid,butyric acid,isovaleric acid,2-Methylbutyric acid,valeric acid,2-methylvaleric acid,4-methylvaleric acid,N-hexanoic acid,and N-heptanoic acid were identified and quantified accurately.Results showed that SCFAs differed significantly between different parts of P.amarus.Main SCFAs were acetic acid,formic acid and propionic acid,basically the same as those in feces.However,the SCFAs in serum were mainly formic acid,acetic acid and propionic acid.We discuss physiological effects of the SCFAs in the panda s intestine and body development.Effects could include maintenance of the balance and integrity of intestinal function of individuals of different ages,and improvement of milk production and body weight of females during lactation.Our study objective was to provide data support for the study of diet selection and physiology of giant panda.
