摘要
动物源性成分鉴定技术在打击肉制品掺假走私、维护市场秩序、保护野生动物资源等方面对检测方法的特异性、灵敏性、重复性提出了很高的要求。在本研究中,首次对黄羊(Procapra gutturosa)及其制品源性成分检测方法进行研究,研究结果表明,普通PCR方法特异性好,与马、牛、羊、猪、鼠、鸡等动物无交叉扩增,灵敏性达到0.1 ng/μL。应用实时荧光定量PCR(Real-time PCR)方法检测特异性好,与马、牛、羊、猪、鼠、鸡等动物无交叉扩增,灵敏性达到0.001 ng/μL,稳定性结果表明,组内试验Ct值的标准差(SD)为0.202,变异系数(CV%)为0.7006%;组间试验Ct值的SD为0.804,CV%为2.845%。黄羊源性成分检测技术方法,具有快速、特异以及灵敏等特点,为准确甄别动物源性成分提供了强有力的技术保障。
The identification of animal-derived ingredients requires high specificity,sensitivity and repeatability of detection methods to combat adulteration and smuggling of meat products,maintain market order,and protect wildlife resources.We studied the detection methods for materials derived from Mongolian gazelle(Procapra gutturosa)for the first time.The results showed that a PCR-based identification approach yielded high specificity.There was no cross amplification with horses,cattle,sheep,pigs,chickens or ducks,and the sensitivity for PCR was 0.1 ng/μL.The sensitivity of real-time fluorescence quantitative polymerase chain reaction(Real-time PCR)was 0.001 ng/μL.Stability test results showed that the coefficient of variation(CV%)of intra-group testing was 0.7%,and that of inter-group testing was 2.8%.This method has the characteristics of rapidity,specificity and sensitivity,which provides a strong technical guarantee for the accurate identification of animal-derived components.
