摘要
目的观察胎盘间充质干细胞(placenta derived mesenchymal stem cells,PDMSCs)移植对阿尔茨海默(Alzheimer's disease,AD)模型大鼠学习记忆能力的影响及其安全性。方法清洁级健康SD雄性大鼠30只,随机数表法分成3组,其中正常组8只、AD模型对照组11只、AD模型移植组11只。采用腹腔注射D-半乳糖制备阿尔茨海默病动物模型。验证AD大鼠造模成功后,AD模型移植组移植细胞悬液浓度为1×105/ml的PDMSCs,AD模型对照组不进行PDMSCs移植,采用同样方法向双侧海马齿状回注射等量生理盐水。移植30 d后进行Morris水迷宫实验,完成定位航行实验和空间探索实验。采集大鼠血样,检测CEA、NSE等肿瘤标志物水平。结果AD模型移植组和AD模型对照组的逃避潜伏期均高于正常对照组的(38.76±19.84)s,但是AD模型移植组逃避潜伏期(54.91±22.36)s低于AD模型对照组的(74.59±27.06)s,组间差异均有统计学意义(P<0.05)。AD模型对照组的第1象限停留时间、穿越平台次数、第1象限活动距离均低于正常对照组和AD模型移植组,组间差异有统计学意义(P<0.05)。AD模型移植组的穿越平台次数、第1象限活动距离与正常对照组差异有统计学意义(P<0.05),但第1象限停留时间组间差异无统计学意义(P>0.05)。AD模型对照组和AD模型移植组两组间NSE、CEA、AFP、CA199、CA125、CA242、F-PSA、Pt、HGH等肿瘤标志物表达差异无统计学意义(P>0.05)。结论胎盘间充质干细胞移植可以改善阿尔茨海默模型大鼠的学习和记忆能力,并且移植后短期无致瘤现象发生,安全性良好。
Objective To observe the effect and safety of placenta derived mesenchymal stem cells(PDMSCs)transplantation on learning and memory ability of Alzheimer's disease(AD)rats.Methods Thirty male SD rats of clean grade were randomly divided into three groups:normal group(n=8),ad model control group(n=11)and AD model transplantation group(n=11).Alzheimer's disease model was established by intraperitoneal injection of D-galactose.PDMSCs with cell suspension concentration of 1×105/ml were transplanted into AD rats.The same amount of saline was injected into bilateral dentate gyrus of hippocampus.Thirty days after transplantation,Morris water maze experiment was carried out to complete positioning navigation experiment and space exploration experiment.Blood samples were collected to detect the levels of CEA,NSE and other tumor markers.Results The escape latencies of AD model transplantation group and AD model control group were higher than those of normal control group(38.76±19.84)s,but the escape latency of AD model transplantation group was(54.91±22.36)s,which was lower than that of AD model control group(74.59±27.06)s.The differences were statistically significant,P<0.05.The length of stay in the first quadrant,the times of crossing the platform,and the activity distance of the first quadrant in the ad model control group were lower than those in the normal control group and AD model transplantation group,and the difference between the two groups was statistically significant(P<0.05).There were significant differences in the times of crossing the platform and the activity distance in the first quadrant between the ad model transplantation group and the normal control group,but there was no significant difference in the first quadrant residence time between the groups,P>0.05.There was no significant difference in NSE,CEA,AFP,CA199,CA125,CA242,f-PSA,Pt,hGH between AD model control group and AD model transplantation group,P>0.05.Conclusion Placental mesenchymal stem cell transplantation can improve the learning and memory ability of Alzheimer's model rats,and there is no tumorigenic phenomenon in a short time after transplantation,with good safety.
