子痫前期患者胎盘蛋白14 与Th17/Treg平衡的相关性研究 被引量:2

摘要 目的:研究子痫前期(PE)患者蜕膜组织中胎盘蛋白14(PP14)与辅助性T细胞17(Th17)、调节性T细胞(Treg)的关系。方法:选取2019年4月至2020年4月在广西乐业县人民医院就诊的PE患者30例作为实验组,同期选取正常妊娠行剖宫产妇女30例作为正常对照组。采用酶联免疫吸附测定(ELISA)法检测蜕膜组织中PP14、白细胞介素(IL)-17和IL-10水平,实时荧光定量PCR(qPCR)法检测蜕膜组织中PP14 mRNA、叉头盒蛋白P3(FOXP3)mRNA、维甲酸相关孤儿受体γ(RORC)mRNA相对表达量。结果:与对照组比较,实验组PP14 mRNA和FOXP3 mRNA相对表达量及PP14、IL-10水平显著降低,RORC mRNA相对表达量和IL-17水平明显升高(均P<0.05)。实验组PP14表达与IL-10、FOXP3呈正相关关系(r=0.730、0.409,P<0.05)。结论:PE患者蜕膜组织中Treg细胞的减少可能与PP14的减少相关,PE母胎界面Th17/Treg细胞的失衡可能与PP14的减少有关。 Objective:To study the relationship between placental protein 14(PP14)and helper T cells17(Th17)and regulatory T cells(Treg)in decidual tissues of preeclampsia(PE)patients.Methods:From April 2019 to April 2020,30 PE patients were selected as experimental group and 30 normal pregnant women were selected as normal control group.The levels PP14,interleukin(IL)-17 and IL-10 in decidual tissues were detected by enzyme linked immunosorbent assay(ELISA),and the mRNA expressions of PP14,forkhead box protein P3(FOX),and retinoic acid receptor-related orphan receptor γ(RORC)were detected by real-time fluorescence quantitative PCR(qPCR).Result:Compared with the control group,The mRNA expressions of PP14 and FOXP3 and the levels of PP14 and IL-10in the experimental group were decreased significantly,while the RORC mRNA expression and IL-17 level were increased(P<0.05).ThePP14 expression was positively correlated with IL-10 and FOXP3 in the experimental group(r=0.730,0.409,P<0.05).Conclusion:The decrease of Treg cells in decidual tissues of preeclampsia patients was related to the decrease of PP14,and the imbalance of Th17/Treg cells in the maternal-fetal interface of PE may be related to the decrease of PP14.
