摘要
目的:探讨异丙酚(PPF)调控Wnt/β-catenin通路对肾癌786-O细胞肿瘤干样特性及运动能力的影响。方法:采用不同剂量(0μmol/L、2.5μmol/L、5μmol/L、10μmol/L)PPF处理786-O细胞。克隆形成法检测细胞增殖,成球实验检测细胞成球直径、成球率,流式分选检测CD133、CD34阳性表达率,Transwell检测细胞侵袭,荧光显微镜观测细胞上皮间充质转化(EMT)形态学,Western blotting法检测血管内皮生长因子(VEGF)、Wnt1、β-连环蛋白(β-catenin)、T细胞因子-4(TCF-4)蛋白表达。结果:与PPF 0μmol/L组比较,PPF 5μmol/L组、10μmol/L组克隆形成率、成球直径、成球率和CD133、CD34阳性表达率以及VEGF、Wnt1、β-catenin、TCF-4蛋白表达水平显著降低,侵袭细胞数显著减少(均P<0.05)。结论:PPF通过抑制Wnt/β-catenin通路降低肾癌786-O细胞增殖、侵袭、EMT和干样特性能力。
Objective:To investigate the effect of propofol(PPF)on cancer stem cell traits and motor ability of kidney cancer 786-O cells through regulation of Wnt/β-catenin pathway.Methods:786-O cells were treated with PPF at different doses 0μmol/L,2.5μmol/L,5μmol/L and 10μmol/L.Cell proliferation was assessed by colony formation assay.The sphere diameter and sphere formation rate were detected by sphere formation assay.The expressions of cancer stem cell markers CD133 and CD34 were detected by flow cytometry.Cell invasion was determined by Transwell assay.The morphology of epithelial mesenchymal transformation(EMT)was observed by fluorescence microscopy.The expressions of vascular endothelial growth factor(VEGF),Wnt1,β-catenin,and T cell factor-4(TCF-4)protein were detected by Western blotting.Results:Compared with PPF 0μmol/L group,5μmol/L and 10μmol/L PPF significantly decreased the colony-forming and sphere formation efficiency,downregulated the expressions of CD133,CD34,VEGF,Wnt1,β-catenin,and TCF-4,and reduced invasive cells(P<0.05).Conclusion:PPF reduced proliferation,invasion,EMT,and cancer stem cell ability of renal carcinoma 786-O cells by inhibition of Wnt/β-catenin pathway.
