摘要
DNA复制起始识别复合物(origin recognition complex,ORC)是一种由六个亚基组成的异构体蛋白复合物,在真核生物DNA复制的启动过程中扮演着重要角色。作为DNA复制的起始蛋白,ORC能够识别复制起始点,通过募集和组装复制起始相关因子协助DNA复制启动。然而,由于DNA复制启动过程的调控机制复杂,并且高等真核生物复制起始序列缺乏特异性,ORC如何准确识别复制起始点并参与DNA复制启动的调控仍未阐明。为此,该文针对真核生物ORC在复制起始中的功能作一综述,以期为相关领域的研究者提供参考。
The origin recognition complex(ORC),a heterogeneous protein complex composed of six subunits,is a core component for eukaryotic DNA replication.As the initiator protein of DNA replication,ORC can identify the replication origin and initiate DNA replication by recruiting and assembling other essential replication factors.However,the regulation mechanism of DNA replication initiation process is complex,and the replication initiation sequence of DNA replication initiation lacks specificity.It is not clear how ORC accurately identifies the replication origin and participates in the regulation of DNA replication initiation.This paper gives a brief review about the roles of ORC in DNA replication initiation process.
