电针对抑郁模型大鼠海马及外周血清BNDF、5-羟色胺的影响 被引量:5

Effect of Electro-Acupuncture on BNDF and 5-HT of Hippocampus and Serum in Treating Depression Model Rats

摘要 目的:通过观察抑郁大鼠的行为学变化、海马内及外周血清脑源性神经营养因子(BDNF)、五羟色胺(5-TH)含量的变化,探讨电针对抑郁状态的治疗作用。方法:将SD大鼠随机分为正常对照组、模型组、电针组和药物组,每组各8只。正常组不接受任何刺激;其余3组采用慢性应激刺激结合孤养建立抑郁模型。模型组不接受任何刺激,电针组每日应激刺激1 h后电针治疗,药物组每日应激刺激1 h后予百忧解灌胃。各组通过高架十字迷宫实验观察大鼠行为学变化,通过免疫组织化学法观察大鼠海马内BDNF及5-TH的表达,酶联免疫吸附法(ELISA)测量大鼠外周血清当中BDNF与5-HT的含量。结果:与正常组相比,模型组大鼠进入开放臂次数比及处于开放臂时间比明显降低(P<0.05),抑郁模型造模成功;与模型组相比,药物组和电针组进入开放臂次数比及时间比明显增加(P<0.05);与药物组相比,电针组大鼠进入开放臂次数比更高(P<0.05)。免疫组化结果显示,与正常组相比,模型组海马BDNF、5-HT表达明显降低(P<0.05);与模型组相比,药物组和电针组BDNF、5-TH表达均明显升高(P<0.05);与药物组相比,电针组中BDNF与5-HT的表达都要稍高。ELISA结果显示,模型组外周血BDNF、5-HT含量明显低于正常组,组间相比差异具有统计学意义(P<0.05);药物组和电针组BDNF、5-HT含量明显高于模型组,组间相比差异具有统计学意义(P<0.05)。结论:抑郁模型大鼠BDNF及5-TH含量显著降低,而电针和抗抑郁药均可以有效提高大鼠海马内及外周血清内BDNF及5-TH含量,改善大鼠抑郁状态及行为活动,且电针效果更优于药物。 Objective:To observe the behavioral changes of rats with depression,the changes of brain-derived neurotrophic factor(BDNF)and 5-hydroxytryptamine(5-TH)in hippocampus and in serum,thus to discuss anti-depression effect of electro-acupuncture.Methods:SD rats were randomly divided into the control group,the model group,the electro-acupuncture group and the medication group,with 8 rats in each group.The depression model was established by chronic stress stimulation combined with isolation.The electro-acupuncture group was treated with electro-acupuncture one hour after stress stimulation,and the medication group was treated with Fluoxetine one hour after stress stimulation.The behavior of rats was observed by elevated plus maze test.The expressions of BDNF and 5-TH in hippocampus were detected by immunohistochemistry,and the contents of BDNF and 5-HT in peripheral blood serum were measured by enzyme-linked immunosorbent assay.Results:Compared to those in the control group,the times that the rats entered the open arm and the duration in the open arm decreased significantly in the model group(P<0.05);after medication intervention and acupuncture intervention,the times and duration were significantly increased compared to those in the model group(P<0.05).Compared to those in the medication group,the times of rats entered the open arm were more in the electro-acupuncture group(P<0.05).The results of immunohistochemistry showed that the depressions of BDNF and 5-HT in hippocampus were significantly lower in the model group than those in the control group(P<0.05);after medication intervention and eletro-acupuncture intervention,they were significantly increased(P<0.05);the increases in the electro-acupuncture group were slightly higher.ELISA results showed that the contents of BDNF and 5-HT in peripheral blood were significantly lower in the model group than those in the control group(P<0.05);after the medication intervention and eletro-acupuncture intervention,the contents were significantly increased(P<0.05).Conclusion:The levels of BDNF and 5-TH in the brain of depression model rats were significantly decreased,whereas electro-acupuncture and antidepressants could effectively increase the contents of BDNF and 5-TH in hippocampus and in serum,improve the depression state and behavioral activities of the rats.The curative effect of electro-acupuncture was better than the medication of Fluoxetine.
