影像学在非人灵长类帕金森模型中的评估进展

摘要 帕金森病(Parkinson's disease,PD)是临床上以运动迟缓、震颤、僵直、姿势不稳为主要表现的常见神经退行性病变,当今医学手段及治疗方法只能延缓其进展却不能完全治愈。成功构建与人类发病机理相似PD实验动物模型是解决这一难题的必要环节。目前PD的动物模型非常多,但非人灵长类(Nonhuman primate,NHP)动物生理、生化机制与人类极其类似,且利用1-甲基-4-苯基-1,2,3,6-四氢吡啶(1-methyl-4-phenyl-1,2,3,6-tet rahydropyridine,MPTP)诱导PD模型在医学研究中的应用非常具有代表性,其代谢性研究也得到普遍认识。影像学的客观和先进性已经在PD模型构建评估时广泛应用,经颅超声、核磁共振、正电子发射断层成像术(PET)和单光子发射计算机断层成像术(SPECT)等都有涉及,但应用情况不一,没有相关文献进行总结,更没有被广泛认可的标准。收集关于NHP动物模型构建的相关文献,从中总结影像学在评估模型构建中的应用情况,综合资料进行比较,以希望总结更加有效的影像学评估手段。建议采用更高磁场(7.0T)的核磁设备及弥散张量成像技术,PET(18F-FP-CIT、[18F]AV-133、8 F-BCPP-EF)及SPECT(99mTc-TRODAT-1)的功能显像技术和视频图像分析系统综合性评估基础实验中帕金森非人灵长类动物模型的构建效果。 Parkinson's disease(PD)is a common neurodegenerative disorder characterized by slow movement,tremor,stiffness and postural instability.However,current medical treatments and treatments can only delay its progression but cannot completely cure it.The successful construction of an experimental animal model of PD similar to human pathogenesis is a necessary step to solve this problem.The animal model of PD is very much,but non-human primates(Nonhuman primate,NHP)animal is very similar with human physiological and biochemical mechanism,and the use of 1-methyl-4-phenyl-6-four hydrogen pyridine(1-methyl-4-phenyl-5-tet rahydropyridine,MPTP)induced PD model application in medical research is very representative,its metabolic research also widely known.The objectivity and advancement of imaging have been widely used in the evaluation of PD model construction,including transcranial ultrasound,nuclear magnetic resonance,positron emission tomography(PET)and single-photon emission computed tomography(SPECT),etc.,but the application is different,there is no summary of relevant literature,and there is no widely recognized standard.Relevant literature on the construction of NHP animal model was collected to summarize the application of imaging in the construction of evaluation model,and comprehensive data were compared to summarize more effective imaging evaluation methods.It is recommended to use nuclear magnetic equipment with higher magnetic field(7.0T)and diffu sion tensor imaging technology,functional imaging technology with PET(18F-FP-CIT、[18F]AV-133、8 F-BCPP-EF)and SPECT(99mTc-TRODAT-1),and video image analysis system to evaluate the effect of building non-human primate mod els of Parkinson's disease in basic experiments.
