康达心口服液对射血分数保留心力衰竭大鼠心肌纤维化的影响 被引量:8

摘要 目的探讨康达心口服液对射血分数保留心力衰竭大鼠心肌纤维化的影响。方法将30只SD大鼠随机分为假手术组(sham组)、射血分数保留心力衰竭组(HFpEF组)和心力衰竭干预组(HFpEF+I组)。采用心脏彩超测量各组大鼠舒张早期二尖瓣血流速度与舒张晚期二尖瓣血流速度比值(E/A)、舒张早期二尖瓣血流速度与舒张早期二尖瓣环运动速度比值(E/e′);采用酶联免疫吸附法检测血清N末端B型钠尿肽原(NT-proBNP)水平;测量并计算心脏重量/总重量比值(Hw/W)和左室重量/总重量比值(LVw/W),同时观察各组大鼠心肌纤维化程度。应用Western Blotting检测各组大鼠心脏组织转化生长因子-β1(TGF-β1)蛋白表达水平。结果与sham组比较,HFpEF组和HFpEF+I组大鼠E/A比值均下降,NT-proBNP水平、E/e′升高,差异均有统计学意义(P<0.05);与HFpEF组比较,HFpEF+I组E/A比值明显升高,NT-proBNP水平、E/e′降低,差异均有统计学意义(P<0.05)。与sham组比较,HFpEF组Hw/W、LVw/W均升高,差异均有统计学意义(P<0.05);与HFpEF组比较,HFpEF+I组Hw/W、LVw/W降低,差异均有统计学意义(P<0.05)。与sham组比较,HFpEF组和HFpEF+I组大鼠心脏组织TGF-β1蛋白表达均升高,差异均有统计学意义(P<0.05);与HFpEF组比较,HFpEF+I组大鼠心脏组织TGF-β1蛋白表达降低,差异有统计学意义(P<0.05)。结论康达心口服液可能通过减少心肌组织TGF-β1蛋白表达水平,抑制心肌纤维化,从而改善射血分数保留心力衰竭大鼠心脏舒张功能。 Objective To explore the effects of Kangdaxin on myocardial fibrosis in Heart failure with preserved ejection fraction rats.Methods Thirty SD rats were randomly divided into 3 groups:sham operation(sham)group,HFpEF(HFpEF)group,HFpEF with drug intervention(HFpEF+I)group.After modeling or treatment,the E/A and E/e′in each group of rats were measured by echocardiography.The level of NT-proBNP was determineded by ELISA.Heart weight/body mass index(Hw/W)and left ventricular weight/body mass index(LVw/W)were accounted after the rats were sacrificed.The expression of TGF-β1 protein in cardiac tissue was detected by Western Blotting.Results Compared with sham group,the E/A in HF group significantly decreased(P<0.05);while the E/e′significantly increased(P<0.05);and the level of NT-proBNP significantly increased(P<0.05).Compared with HF group,the value of E/A in HFpEF+I group significantly increased(P<0.05);while the E/e′significantly decreased(P<0.05);and the level of NT-proBNP significantly decreased(P<0.05).Compared with sham group,Hw/W and LVw/W in HFpEF group significantly increased(P<0.05).Compared with HFpEF group,Hw/W and LVw/W in HFpEF+I group significantly decreased(P<0.05).Compared with sham group,the expression of TGF-β1 protein in heart tissue of HFpEF group significantly increased(P<0.05),compared with HFpEF group,the expression of TGF-β1 protein in HFpEF+I group significantly decreased(P<0.05).Conclusion Kangdaxin oral liquid had protective effect on heart in HFpEF rats,which could reduce the protein expression of TGF-β1 in the heart tissue of HFpEF rats.Its mechanism might be to inhibit myocardial fibrosis and improve cardiac diastolic function.
