滋膵通脉饮对2型糖尿病大鼠糖脂代谢和胰岛素抵抗的影响 被引量:3

Effect of Zicui Tongmai Decoction on glycolipid metabolism and insulin resistance in rats with type 2 diabetes mellitus

摘要 目的探讨滋膵通脉饮对2型糖尿病大鼠糖脂代谢和胰岛素抵抗的影响。方法取45只SD大鼠,随机抽取10只作为空白对照组,剩余35只采用高脂高糖饲料喂养+腹腔注射链脲佐菌素的方法构建2型糖尿病大鼠模型。将造模成功的30只2型糖尿病大鼠随机分模型组、滋膵通脉饮组和罗格列酮组,每组10只。于实验第7周开始,滋膵通脉饮组大鼠灌服滋膵通脉饮(生药量为17.6 g/kg),罗格列酮组大鼠灌服罗格列酮混悬液[0.42 mg/(kg·d)],其他两组大鼠均灌服同等体积的蒸馏水。4周后检测4组大鼠血清胆固醇、三酰甘油、LDL、空腹血糖、空腹胰岛素水平,计算稳态模型胰岛素抵抗(HOMA-IR)指数。结果与空白对照组比较,其余各组空腹血糖、空腹胰岛素和HOMA-IR指数,总胆固醇、三酰甘油和LDL水平均升高(均P<0.05)。滋膵通脉饮组和罗格列酮组空腹血糖、空腹胰岛素和HOMA-IR指数均低于模型组(均P<0.05),但两组间差异无统计学意义(均P>0.05)。滋膵通脉饮组胆固醇、三酰甘油和LDL水平低于模型组,且三酰甘油水平低于罗格列酮组(均P<0.05);而罗格列酮组和模型组之间以上血脂指标水平差异无统计学意义(均P>0.05)。结论滋膵通脉饮能降低2型糖尿病大鼠血糖、改善胰岛素抵抗,效果与罗格列酮相当,同时其还能调节血脂紊乱。 Objective To explore the effect of Zicui Tongmai Decoction on glycolipid metabolism and insulin resistance in rats with type 2 diabetes mellitus(T2DM).Methods Among 45 SD rats,10 rats were randomly selected as blank control group,and the remaining 35 rats were modeled as T2DM rats by high-fat and high-glucose diet and intraperitoneal injection with streptozotocin.Then the 30 T2DM rats with successful modeling were randomized to model group,Zicui Tongmai Decoction group or rosiglitazone group,with 10 rats in each group.At the 7th week of the experiment,rats in the Zicui Tongmai Decoction and rosiglitazone groups were treated with Zicui Tongmai Decoction(with a crude drug amount of 17.6 g/kg)and rosiglitazone(0.42 mg/(kg·d))by gavage,respectively,rats in the other two groups were administered isometric distilled water by gavage.After four weeks,the levels of serum cholesterol,TG,LDL,FBG and fasting insulin of rats were determined in the four groups,and homeostasis model assessment for insulin resistance(HOMA-IR)index was calculated.Results By comparison with the blank control group,the remaining groups yielded increased levels of FBG,fasting insulin,HOMA-IR index,TC,TG and LDL(all P<0.05).FBG,fasting insulin and HOMA-IR index in the Zicui Tongmai Decoction and rosiglitazone groups were lower than those in the model group(all P<0.05),but no statistically significant difference was found between the former two groups(all P>0.05).The Zicui Tongmai Decoction group obtained lower cholesterol,TG and LDL levels as compared with the model group,and also exhibited declined TG level as compared with the rosiglitazone group(all P<0.05);but there was no statistically significant difference in aforementioned blood lipid indicators levels between the rosiglitazone group and the model group(all P>0.05).Conclusion Zicui Tongmai Decoction can not only exert a similar effect on reducing blood glucose and improving insulin resistance as rosiglitazone,but also regulate blood lipid disorder in T2DM rats.
