摘要
目的探讨益生菌对重症急性胰腺炎(SAP)大鼠肠黏膜的保护作用及回肠差异表达蛋白的影响。方法将24只健康雄性大鼠随机分为假手术组(SO组)、ANP组、PRO组,每组8只。ANP组及PRO组构建SAP模型,PRO组建模后给予益生菌灌胃,ANP组及SO组给予等量生理盐水灌胃。比较3组建模后24 h及48 h的血淀粉酶水平、建模后48 h胰腺及回肠病理组织学及评分。分析回肠差异表达蛋白及其生物学功能。结果(1)造模后24 h、48 h SO组、PRO组、ANP组血清淀粉酶水平均依次升高;造模后48 h,ANP组和PRO组胰腺病理评分高于SO组。ANP组回肠组织病理评分高于SO组、PRO组(P<0.05)。(2)ANP组与SO组间共筛选出49个差异表达蛋白,其中12个蛋白质表达上调,37个蛋白质表达下调,PRO组与SO组间共筛选出107个差异表达蛋白,其中6个蛋白质表达上调,101个蛋白质表达下调。其中胶转蛋白、半乳糖凝集素-1是与SAP发生后肠黏膜损伤关联最强的差异蛋白。结论益生菌能减轻SAP大鼠肠黏膜损伤,其可能通过差异表达蛋白的表达而发挥作用。
Objective To explore the protective effect of probiotics on intestinal mucosa and its impact on differentially expressed ileum protein in rats with severe acute pancreatitis(SAP).Methods Twenty-four healthy male rats were randomly divided into sham operation group(SO group),ANP group and PRO group,with eight rats in each group.SAP model was constructed in the ANP and PRO groups,the PRO group was given probiotics by gavage after modeling,the ANP and SO groups were given the same amount of normal saline by gavage.Serum amylase levels 24 and 48 hours after modeling,and histopathology and scores of the pancreas and ileum 48 hours after modeling were compared among the three groups.Differentially expressed ileum protein and its biological function were analyzed.Results(1)Twenty-four and 48 hours after modeling,serum amylase levels in the SO group,the PRO group and the ANP group increased in turn;48 hours after modeling,the pathological scores of the pancreas in the ANP and PRO groups were higher than those in the SO group.The pathological scores of ileum tissues in the ANP group were higher than those in the SO and PRO groups(P<0.05).(2)A total of 49 differentially expressed proteins were screened out between the ANP group and the SO group,including 12 proteins with up-regulated expression and 37 proteins with down-regulated expression.A total of 107 differentially expressed proteins were screened out between the PRO group and the SO group,including six proteins with up-regulated expression and 101 proteins with down-regulated expression.Among them,transgelin and galectin-1 were the differentially expressed proteins which showed the most strong correlation with post-SAP intestinal mucosa injury.Conclusion Probiotics can ameliorate intestinal mucosa injury in SAP rats,which plays a role probably by the expression of differentially expressed protein.
