基于网络药理学探讨鼻咽解毒胶囊治疗鼻咽癌的作用机制

Mechanism of Biyan Jiedu Capsule on Nasopharyngeal Carcinoma Based on Network Pharmacology

目的:基于网络药理学方法探讨鼻咽解毒胶囊抗鼻咽癌的作用机制。方法:通过TCMSP数据库挖掘鼻咽解毒胶囊活性成分,查阅相关文献并限定口服生物利用度(OB)和类药性(DL)数值对鼻咽解毒胶囊活性成分进行筛选并获取靶点,通过Uniprot数据库查询靶点对应的基因,采用Cytoscape软件构建鼻咽解毒胶囊活性成分-靶点、靶点-疾病网络;通过DAVID数据库对鼻咽解毒胶囊抗鼻咽癌作用靶点进行基因本体(GO)功能分析和KEGG通路分析,以探究鼻咽解毒胶囊抗鼻咽癌的作用机制。结果:筛选得到33个潜在活性成分和131个潜在作用靶点;化合物-靶点网络显示,关键成分包括槲皮素、山奈酚、芒柄花黄素、汉黄芩素等,关键靶点包括NR3C4、ESR1、PTGS2、PIM1、PPARG、NOS2等。GO功能富集分析得到GO条目160个,其中生物过程(BP)条目100个,分子功能(MF)条目43个,细胞组成(CC)条目17个;主要涉及蛋白结合、细胞、酶结合等;KEGG富集得到45条代谢通路,包括癌症通路、EB病毒感染、血管内皮生长因子(VEGF)等与鼻咽癌密切相关的通路,还涉及内分泌系统、免疫系统等多条与肿瘤形成、发展相关的通路。结论:鼻咽解毒胶囊治疗鼻咽癌具有多成分、多靶点、多途径的作用特点,其具体的调控机制及关键的靶标尚待通过进一步的实验研究加以验证。

Objective:To explore the mechanism of Biyan Jiedu Capsule in the treatment of nasopharyngeal carcinoma based on network pharmacology.Methods:TCMSP was used to search for the chemical constituents of biyan Jiedu Capsules,relevant literature consult and the oral bioavailability(OB)and drug-like(DL)values were used to screen the active ingredients of Biyan Jiedu Capsules and obtaining targets,querying through Uniprot database Genes corresponding to the target,the active ingredient-target network and target-disease network was constructed by Cytoscape software;The mechanism of its action was studied by DAVID database analyzing enrichments of gene ontology(GO)function and pathway enrichments of KEGG.Results:33 potential active ingredients and 131 potential targets were screened;the compound-target network showed that the key ingredients include quercetin,kaempferol,formononetin,and Wogonin,and the key targets include NR3C4,ESR1,PTGS2,PIM1,PPARG,NOS2,etc.GO functional enrichment analysis yielded 160 GO entries,including 100 biological process(BP)entries,43 molecular function(MF)entries,and 17 cell composition(CC)entries;mainly involving protein binding,cells,and enzyme binding;KEGG enriches 45 metabolic pathways,including cancer pathways,Epstein-Barr virus infection,vascular endothelial growth factor(VEGF)and other pathways that are closely related to nasopharyngeal carcinoma,and also involve the endocrine system,immune system and other pathways related to tumor formation and development Access.Conclusions:Biyan Jiedu Capsules has the characteristics of multi-component,multi-target and multi-path effects in the treatment of nasopharyngeal carcinoma.However,the specific regulation mechanism and key targets need to be verified by further experimental studies.
