2型囊泡单胺转运体分子探针18F-FP-(+)-DTBZ纵向定量评价2型糖尿病大鼠脑多巴胺能神经元

摘要 目的观察2型囊泡单胺转运体(VMAT2)靶点分子探针18F-FP-(+)-DTBZ评价2型糖尿病(T2DM)大鼠脑多巴胺能神经元的效果。方法将12只ZDF大鼠均分为T2DM组(fa/fa基因型)和对照组(fa/+基因型),每组6只。于大鼠3、6和12月龄时行18F-FP-(+)-DTBZ Micro-PET/CT显像,观察2组大鼠体质量、血糖及脑纹状体标准摄取比值(SUVR)变化。结果 T2DM组大鼠6月龄时体质量明显高于对照组(P<0.05),而3月和12月龄时2组体质量差异均无统计学意义(P均>0.05)。T2DM组大鼠3月龄时空腹血糖与对照组差异无统计学意义(P>0.05),而6月龄、12月龄时空腹血糖均明显高于对照组(P均<0.05)。T2DM组大鼠3月龄、6月龄时脑纹状体SUVR与对照组差异均无统计学意义(P均>0.05),12月龄时明显低于对照组(P<0.05)。结论 2型糖尿病可致大鼠脑纹状体VMAT2表达降低,进而引起脑多巴胺能神经元功能异常。 Objective To observe the effect of 18 F-FP-(+)-DTBZ,vesicular monoamine transporter 2(VMAT2)molecular probe for evaluating dopaminergic neurons in type 2 diabetes mellitus(T2DM)rats.Methods Twelve ZDF rats were equally divided into T2DM group(fa/fa)and the control group(fa/+)(each n=6).18 F-FP-(+)-DTBZ Micro-PET/CT imaging was performed when the rats being 3,6 and 12-month old in both groups to observe the changes of rats'mass,glucose and the standardized uptake value ratio(SUVR)in the striatum of brains.Results The rats'mass of those being 6-month old in T2DM group was significantly higher than that in control group(P<0.05),while there was no significant difference between groups for rats being 3 and 12-month old(both P>0.05).The fasting blood glucose of rats being 3-month old was not significant different between groups(P>0.05),while of rats being 6 and 12-month old were significantly higher in T2DM group than in control group(both P<0.05).The striatal SUVR in T2DM group was significantly lower than that of control group for rats being 12-month old(P<0.05),but there was no significant difference between groups for rats being 3 and 6-month old(both P>0.05).Conclusion T2DM could result decrease of VMAT2 expression in the striatum of rat's brain,which might lead to abnormal function of brain dopaminergic neurons.
