白花丹素对卵巢癌SKOV3细胞增殖和凋亡能力的影响情况研究

Study on the effect of Baihuadan on the proliferation and apoptosis of ovarian cancer SKOV3 cells

目的:探究白花丹素对卵巢癌SKOV3细胞的增殖和凋亡能力的影响情况。方法:将细胞分为对照组、PB1组、PB2组和PB5组,各组加入相应浓度的白花丹素处理细胞,CCK8检测细胞增殖,Hoechst检测细胞凋亡,Western blot检测蛋白表达。结果:PB1组、PB2组和PB5组Ki67、Bcl-2蛋白表达水平、增殖能力均显著低于对照组,且变化趋势同白花丹素浓度呈负相关,差异有统计学意义(P<0.05);同时,细胞凋亡、Bax、cleaved Caspase-9蛋白表达水平显著升高,变化趋势同白花丹素浓度呈正相关,差异有统计学意义(P<0.05)。结论:白花丹素能促进癌细胞凋亡和抑制卵巢癌细胞的增殖,作用机制可能与蛋白表达有关。

Objective:To investigate the effect of Baihuadan on the proliferation and apoptosis of ovarian cancer SKOV3 cells.Methods:The cells were divided into the control group,the PB1 group,the PB2 group and the PB5 group.The cells were treated with corresponding concentration of Baihuadan.The cell proliferation was detected by CCK8 and the expression of apoptosis protein was detected by Hoechst,detection of protein expression by Western blot.Results:The expression level of Ki67 and Bcl-2 protein and the ability of proliferation in the PB1 group,the PB2 group and the PB5 group were significantly lower than those in the control group,and the change trend was negatively correlated with the concentration of blephardanthin,the differences were statistically significant(P<0.05).At the same time,the expression of apoptosis,Bax and cleaved Caspase-9 protein increased significantly,and the change trend was positively correlated with the concentration of blephardanthin,the differences were statistically significant(P<0.05).Conclusion:Baihuadan can promote the apoptosis of ovarian cancer cells and inhibit the proliferation,and the mechanism may be related to the expression of protein.
