化瘀通络灸对VD大鼠体外干预HUVEC-12细胞VEGF、bFGF蛋白表达的影响 被引量:3

Effect of Huayu Tongluo Moxibustion on the expression of VEGF and bFGF protein in HUVEC-12 cells of VD rats in vitro

摘要 目的观察化瘀通络灸对VD大鼠血清体外干预HUVEC-12细胞VEGF、bFGF蛋白表达的影响。方法采用四血管阻断法制造VD大鼠模型,随机分为艾灸组、模型组、西药组,另设假模型组作为对照,艾灸组采用化瘀通络灸法,取百会、神庭、大椎艾灸,西药组茴拉西坦灌胃,每日一次,治疗2个疗程,采用跳台实验检测各组神经行为学评分。取艾灸组、模型组、假模型组VD大鼠的血清体外培养HUVEC-12细胞,另设平行培养液组作为对照,采用Western blot法检测各组VEGF、bFGF的蛋白表达。结果治疗结束后,与模型组潜伏期为(36.25±10.71)s、错误次数为(6.19±0.74)次、神经行为学评分为(3.00±0.53)分比较,艾灸组和西药组的潜伏期为(232.31±22.25)s和(201.38±13.12)s,均延长(P<0.01);错误次数为(1.00±0.56)次和(2.53±0.84)次及神经行为学评分为(1.00±0.56)分和(1.74±0.47)分,均降低(P<0.01)。艾灸组和西药组比较,差异有统计学意义(P<0.05)。与模型组、假模型组、平行液组比较,艾灸组的HUVEC-12细胞VEGF、bFGF的蛋白表达水平(0.84±0.09)和(0.87±0.05)较高,有显著差异(P<0.01)。结论化瘀通络灸法能够明显提高VD大鼠学习记忆能力,增强HUVEC-12的VEGF、bFGF的蛋白表达,其治疗VD可能与其促血管生成作用相关,为针灸治疗VD提供理论依据。 Objective To observe the effect of Huayu Tongluo Moxibustion on the expression of VEGF and bFGF protein in HUVEC-12 cells of VD rats in vitro.Methods The four-vessel blocking method was used to make VD rat models,and they were randomly divided into the moxibustion group,model group,and western medicine group.Another fake model group was set up as the control group.The moxibustion group was treated with moxibustion for removing blood stasis and dredging collateral,taking Baihui and Shenting,Dazhui for moxibustion.The western medicine group was given aniracetam lavage once a day for two courses of treatment.The platform jumping experiment was used to detect the neurobehavioral score of each group.HUVEC-12 cells were cultured in vitro from the serum of VD rats in the moxibustion group,the model group,and the sham model group,and the parallel solution group was also set up as the control group.Western blot was used to detect the protein expression of VEGF and bFGF in each group.Results After treatment,compared with the model group's incubation period was(36.25±10.71)seconds,the number of errors was(6.19±0.74)times,and neurobehavioral score was(3.00±0.53)points,the incubation period of the moxibustion group and the western medicine group was(232.31±22.25)seconds and(201.38±13.12)seconds were prolonged(P<0.01);The number of errors was(1.00±0.56)times and(2.53±0.84)times and neurobehavioral scores was(1.00±0.56)points and(1.74±0.47)points were decreased(P<0.01).The difference between the moxibustion and western medicine groups was statistically signifi cant(P<0.05).Compared with those of the model group,sham model group,and parallel solution group,the protein expression levels of VEGF and bFGF in HUVEC-12 cells in the moxibustion group([0.84±0.09]and[0.87±0.05])were higher,with significant differences(P<0.01).Conclusion Huayu Tongluo Moxibustion can significantly improve the learning and memory ability of VD rats,and enhance the protein expression of VEGF and bFGF of HUVEC-12.Its treatment of VD may be related to its pro-angiogenesis effect,which provides a theoretical basis for VD's acupuncture treatment.
