岷县当归根腐病病原菌的分离鉴定及生物学特性研究 被引量:10

摘要 为探究当归根腐病病害的主要病原菌及其生物学特性,本研究选取患根腐病的岷县当归为材料,进行了病原菌的分离纯化、菌株形态学观察、分子生物学鉴定等试验,同时分析了不同碳源、氮源、温度、pH、光照对病原菌生长的影响,以及菌丝的致死温度.结果表明,岷县当归根腐病病原菌为燕麦镰刀菌(Fusarium avenaceum)和锐顶镰刀菌(Fusarium acuminatum),燕麦镰刀菌在以可溶性淀粉、葡萄糖和蔗糖为碳源,蛋白胨为氮源,温度为20℃,pH值为8时生长较好,光照对燕麦镰刀菌的生长无明显影响,63℃处理10 min可致死;锐顶镰刀菌在以甘露醇、葡萄糖和蔗糖为碳源,蛋白胨为氮源,温度为25℃,pH值为8,连续黑暗或12 h光暗交替培养下生长较好,71℃处理10 min可致死. In order to explore the main pathogens and biological characteristics of root rot disease of Angelica sinensis,diseased A.sinensis sample was selected as the experimental material to isolate and identify the pathogen.The effects of different carbon sources,nitrogen sources,temperature,pH,and light on the growth of pathogen and the lethal temperature of mycelium were analyzed.The results showed that the pathogens of A.sinensis root rot in Minxian County were Fusarium avenaceum and F.acuminatum.F.avenaeum grew better by using soluble starch,glucose and sucrose as carbon source,peptone as nitrogen source,at 20℃and in pH value of 8,and light had no obvious effect on the growth of F.avenaeum.Treatment at 63℃for 10 minutes could make it inactive.And the F.acuminatum grew better by using mannitol,glucose and sucrose as carbon source,peptone as nitrogen source,at 25℃and in pH value 8 and continuous darkness or 12 h light and dark alternating culture,treatment at 71℃for 10 minutes could make it inactive.
