红叶石楠耐铅内生细菌的分离及其16S rDNA鉴定 被引量:1

摘要 通过生理分析和分子鉴定相结合的方法,对种植于公路两旁的红叶石楠根茎叶不同部分的内生菌进行分离纯化。结果表明,从不同组织中初步分离出内生细菌44株,均为革兰氏阳性菌,对这些菌株做生理生化鉴定;提取生理生化结果有差异的细菌DNA并扩增其16S rDNA序列,通过BLAST比对分析并利用MEGA软件建立发育树等分子鉴定手段初步推断,这些内生细菌在亲缘关系上与不动杆菌属、假单胞菌属和蜡样芽孢杆菌最为接近。本研究为铅污染地区土壤的微生物与植物联合的生物修复提供了行之有效的技术理论基础。 The method of identifying the physiological analysis and molecular identification were involved in this study.Then isolation and purification of endophytic Bacteria from different parts of Photinia serrulata were carried out.The results showed that 44 strains endophytic bacteria isolated and purified were all Gram-positive bacteria,and 44 strains endophytic bacteria were identified physiologically and biochemically.DNA was extracted from bacteria,16 S r DNA sequence by PCR,blast analysis,phylogenetic tree using establishing by MEGA software.It was inferred that in terms of kinship,these endophytic bacteria had a close relation with Pseudomonas,Bacillus cereus and Acinetobacter.This study provided an effective technical basis for microbial-plant combined bioremediation technology in lead contaminated areas.
