家蚕血淋巴多肽鉴定与分析

摘要 多肽在生命活动中扮演着非常重要的角色,几乎参与所有生理过程。本研究以家蚕5龄第3天血淋巴进行多肽组测序,以高效液相色谱-质谱法(LC-MS/MS)分离多肽并进行质谱鉴定,最终鉴定到295个多肽,长度主要集中在10~35个氨基酸残基,功能聚类分析表明主要为载脂蛋白apolipophorin-Ⅲ、抗菌肽、gloverin、Y-box相关蛋白等,此外还鉴定到大量未知多肽,这些多肽有些是功能性多肽,有些可能是前体蛋白的降解产物。本研究对家蚕血淋巴进行多肽组测序,为昆虫多肽鉴定和功能分析提供了实验证据,对多肽在生理活动中的作用,尤其是抗菌肽在昆虫先天性免疫中的作用研究具有参考价值。 Peptides play irreplaceable roles throughout almost all physiological activities in the life. In this study, peptidomic analysis on the hemolymph of 3 rd day 5 th instar silkworm(Bombyx mori) larva was performed using liquid chromatography-mass spectrometry(LC-MS/MS). Consequently, 295 peptides were identified, and their lengths were primarily 10 to 35 amino acid residues. Gene ontology analysis exhibited that they were apolipophorin-Ⅲ, antimicrobial peptide, gloverin, Y-box related proteins in addition to many unknown peptides. Among them, some were functional peptides while others might be degradation products of precursor proteins. The results provide experimental evidence for peptide identification and functional analysis based on peptidomic analysis on silkworm hemolymph. Moreover, it facilitates the study on the function of peptides in the physiological activity, especially on the role of antimicrobial peptides in innate immunity of insect.
