摘要
利用超高效液相色谱(UPLC)法建立同时测定紫菀药材中绿原酸,异绿原酸A,异绿原酸B含量的方法。方法:采用UPLC法,Acquity UPLC BEH C18色谱柱(2.1 mm 50 mm,1.7μm);乙腈-0.1%磷酸水溶液为流动相梯度洗脱;流量:0.20 mL/min;检测波长:330 nm;柱温:30℃;结果:绿原酸、异绿原酸A、异绿原酸B分别在0.463~46.3μg、0.4541~45.41μg、0.1430~14.30μg范围内线性关系良好。加样回收率分别为:绿原酸回收率为98.7%(RSD=1.68%),异绿原酸A的回收率为96.6%(RSD=1.04%),异绿原酸B的回收率为97.14%(RSD=0.98%)、准确度高、稳定性良好、重现性较好。运用此方法测定亳州产紫菀不同地下部位的绿原酸类物质的含量,实验得出根茎中绿原酸类物质含量最高,其他部位绿原酸、异绿原酸A、异绿原酸B含量较低。结论:该方法灵敏度高,快速敏捷,重复性好,可以作为研究紫菀药材中绿原酸类物质质量的依据。
The paper establishes a method for the simultaneous determination of chlorogenic acid,isochlorogenic acid A and isochlorogenic acid B in Aster by ultra-high performance liquid chromatography(UPLC).Methods:UPLC,Acquity UPLC BEH C18 column(2.1 mm 50 mm,1.7μm)is used,the mobile phase is eluted with acetonitrile-0.1%phosphate aqueous solution,the flow rate is 0.20 mL/min,the detection wavelength is 330 nm,the column temperature is 30℃,the flow rate is 0.20 mL/min,the detection wavelength is 330 nm,and the column temperature is 30℃.Results:The linear relationship is good when chlorogenic acid,isochlorogenic acid A and isochlorogenic acid B in the range of 0.463μg^46.3μg,0.4541μg^45.41 and 0.1430~14.30μg respectively.The recovery rates of chlorogenic acid,isochlorogenic acid A and isochlorogenic acid B are 98.7%(RSD=1.68%),96.6%(RSD=1.04%)and 97.14%(RSD=0.98%)respectively.The stability is good and the reproducibility is good.The content of chlorogenic acids in different underground parts of Aster Bozhou is determined by this method.The results show that the content of chlorogenic acids in roots and stems is the highest,and that in other parts is the highest.The contents of isochlorogenic acid A and isochlorogenic acid B are lower.Conclusion:the method is accurate,reliable,sensitive,rapid and reproducible,and can be used as a basis for the study of chlorogenic acids in Aster.
