摘要
为优化烟草花药培养体系,以♂净叶黄×♀HKDN-5的杂交子一代为材料,采用响应面法考察了IAA浓度、6-BA浓度和大量元素浓度3个因素对烟草花药胚状体诱导的影响。结果表明:烟草花药胚状体诱导的最佳条件为0.49 mg/L IAA+0.48 mg/L 6-BA+0.56 MS,胚状体的诱导率为36.80%。为了便于实际操作,可采取0.50 mg/L IAA+0.50 mg/L 6-BA+1/2 MS的条件进行试验。
Response surface methodology(RSM)was used to optimize the anther culture system by using the hybrid progeny of♂Jingyehuang×♀HKDN-5 as the material.According to the RSM design,the tobacco anther were treated by 17 combinations which consisted of IAA,6-BA and macroelements.The results of anther culture test such as anther embryoid induction rate and callus induction rate were measured and analyzed by response surface analysis.The optimal combination was 0.49 mg/L IAA+0.48 mg/L 6-BA+0.56 MS,obtaining an embryoid induction rate of 36.80%;for practical convenience,it can take 0.50mg/L IAA+0.50mg/L 6-BA+1/2 MS.
