基于网络药理学探讨温中益气方抗胃溃疡的作用机制

Action Mechanism of Wenzhong Yiqi Formula in Treating Gastric Ulcer Based on Network Pharmacology

目的:基于网络药理学的研究方法探讨温中益气方抗胃溃疡作用机制。方法:通过检索TCMSP、DrugBank、OMIM、GeneCard等数据库,查找确认温中益气方活性成分、作用靶点以及胃溃疡相关靶点,并通过STRING分析各靶点之间的相互作用。利用Cytoscape3.7.2软件进行药物-成分-疾病-靶点网络分析,在此基础上采用R3.5.2(依托于“clusterProfiler”“org.Hs.eg.db”以及“pathview”程序包)进行GO与KEGG基因功能富集分析。结果:通过筛选后,最终获得温中益气方233个活性成分,125个作用靶点,与胃溃疡相关靶点95个。根据GO功能富集分析显示,温中益气方抗胃溃疡的作用机制主要集中于多种细胞核因子调控,细胞内氧化还原反应与氧化应激损伤,泛素/泛素样蛋白连接酶系统,NF-κB结合,RNA聚合酶II近端启动子序列等多个细胞内生命活动密切相关。KEGG功能富集分析主要集中NF-κB等信号通路,主要与炎症、细胞内应激反应、自噬、细胞凋亡、纤维化信号通路、抗氧化应激,白细胞募集等密切相关。结论:温中益气方在治疗胃溃疡过程中是以NF-κB信号通路为主,多种信号通路联合协同作用。

Objective:To explore the action mechanism of Wenzhong Yiqi Formula in treating gastric ulcer(GU).Methods:Databases of TCMSP,DrugBank,OMIM and GeneCard were searched to identify the active components,targets and GU related targets of Wenzhong Yiqi Formula,and the interaction among the targets was analyzed by STRING.The drug-composition-disease-target network was analyzed by using the software of Cytoscape 3.7.2.On this basis,R3.5.2,which relied on package of"clusterProfiler","org.Hs.eg.db"and"pathview",were used to analyze the gene functional enrichment analysis of GO and KEGG.Results:After screening,233 active components,125 targets and 95 GU related targets were obtained.According to GO functional enrichment analysis,the mechanism of Wenzhong Yiqi Formula in treating GU mainly focused on the regulation of various nuclear factors.Intracellular redox reaction was closely related to oxidative stress injury,ubiquitin/ubiquitin like protein ligase system,NF-κB binding and RNA polymerase II proximal promoter sequence.KEGG functional enrichment analysis mainly focused on NF-κB signaling pathway,which was closely related to inflammation,intracellular stress response,autophagy,apoptosis,fibrosis signaling pathway,antioxidant stress and leukocyte recruitment.Conclusion:Wenzhong Yiqi Formula is mainly involved in NF-κB signaling pathway in the treatment of GU.
