摘要
目的探讨蒙药色日古德(长松萝)醇提物的正丁醇萃取成分(简称示例药物)对D-氨基半乳糖(D-GalN)中毒大鼠急性肝损伤的解毒作用机理。方法以D-GalN诱导建立大鼠急性肝损伤模型,观察示例药物对血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、白蛋白(ALB2L)、总蛋白(TP2)、碱性磷酸酶(ALP2L)、甘油三酯(TG)、胆碱酯酶(CHE2)活性变化。取固定于20%甲醛溶液的肝组织、利用HE染色法观察肝组织病理变化。并利用免疫组化法观察肝细胞的Bcl-2表达的影响。结果示例药物A、B组明显降低清ALT、AST和升高ALB2、TP2活性;生药组有降低ALP2L、TG活性作用。结论蒙药长松萝正丁醇层对D-GalN中毒肝脏具有保护作用。
Objective To investigate the detoxification mechanism on the acute liver injury induced by D-galactose(D-GalN)in Mongolian medicine Usnea longissima Ach.Methods Rat model of acute liver injury was induced by D-GalN.The activities of serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),albumin(ALB2L)and total protein(TP2),alka-line phosphatase(ALP2L),triglyceride(TG),cholinesterase(CHE2)were observed.The liver tissue fixed in 20%formalde-hyde solution was taken and the pathological changes of liver tissue were observed by HE staining.The effect of Bcl-2 expression in hepatocytes was observed by immunohistochemistry.Results The exemplified drugs A and B significantly reduced ALT,AST and increased ALB2 and TP2 activities;the crude drug group decreased ALP2L and TG activities.Conclusion The Mongolian medicine Usnea longissima Ach.butanol layer has protective effect on D-GalN poisoning liver.
