摘要
目的探讨小儿腹泻外敷凝胶贴膏对大鼠小肠绒毛及血清和小肠组织中主要神经递质的影响。方法Wistar大鼠随机分成5组,空白组、模型组、药物高、中、低剂量组,每组10只,采用20%的番泻叶水煎液灌胃(2ml/100g)建立腹泻模型。采用HE染色法观察小儿腹泻外敷凝胶贴膏对大鼠肠绒毛的影响;采用ELISA法检测敷药前后大鼠血清中一氧化氮(NO)含量变化,采用放射免疫法测定小肠组织中P物质(SP)、血管活性肠肽(VIP)、乙酰胆碱(ACH)含量的变化。结果小儿腹泻外敷凝胶贴膏对大鼠小肠绒毛具有良好的修复作用;同时可以提高血清中NO、小肠组织中VIP的含量,降低小肠组织中SP、ACH的含量。结论小儿腹泻外敷凝胶贴膏可以通过改善小肠绒毛功能,降低兴奋性神经递质含量,提高抑制性神经递质的含量来改善番泻叶所致的大鼠腹泻。
Objective To study the effect of Xiaoer Fuxie Waifu gel emplastrum on intestinal villi,and major neurotransmitters in serum and small intestine of rats.Methods Wistar rats were randomly divided into 5 groups:blank group,model group,high-dose,medium-dose and low-dose groups,ten rats in each group.The diarrhea model was established by gavage 20%water decoction of senna leaf(2ml/100g).HE staining method was used to observe the effect of Xiaoer Fuxie Waifu gel emplastrum on in-testinal villi of rats.The changes of nitric oxide(NO)in serum of rats were detected by ELISA,and the changes of substance P(SP),vasoactive intestinal peptide(VIP)and acetylcholine(ACH)in small intestine tissues were detected by radioimmunoas-say.Results The Xiaoer Fuxie Waifu gel emplastrum had a good effect on the repair of intestinal villi in rats.At the same time,the concentration of NO in serum and VIP in small intestine could be increased and the concentration of SP and ACH in intestinal tissues could be reduced.Conclusion Xiaoer Fuxie Waifu gel emplastrum can improve diarrhea of rats induced by senna leaf by improving villi function,reducing the concentration of excitatory neurotransmitter and increasing the concentration of inhibitory neurotransmitter.
