人诱导性多能干细胞及分化产物在发育毒性研究中的应用

摘要 个体成年之前因接触外源化学物质而产生的任何有害影响均会造成自身生长发育异常,因此,药物开发乃至临床运用均需要可靠的发育毒性实验数据来证明其可用性。人诱导性多能干细胞(hiPSCs)作为一种新型的体外细胞来源,其生理特点与胚胎干细胞类似,具有无限增殖和多向分化的潜能。近年来,不断有研究检测了外源物对hiPSCs及其衍生的各种成体细胞的毒性作用,除了对生长形态的影响外,还尝试探索了对生理功能及各种特异标志物等的影响及可能机制,以期为药物发育毒性评价提供一种简单、可再生、经济有效的体外细胞替代模型。 Any harmful effects of exposure to chemicals before adulthood can result in abnormal growth and development,so the development and clinical application of a drug require reliable experimental data of developmental toxicity to prove its feasibility.As a new type of in vitro cell source,human induced pluripotent stem cells(hiPSCs)have similar physiological characteristics to embryonic stem cells and have the potential for infinite proliferation and multi-directional differentiation.In recent years,there have been continuous studies to detect the toxic effects of drugs on hiPSCs and their derived adult cells.In addition to the effects on growth morphology,the effects and possible mechanisms on physiological functions and various specific markers have also been tried to be explored,in order to provide a simple,reproducible,and cost-effective in vitro cell replacement model for the evaluation of developmental toxicity of drugs.
