摘要
核因子E2相关因子2(Nrf2)是一种细胞内转录调节因子,血红素加氧酶1(HO-1)是其最重要的下游调控产物之一,两者的级联反应在机体抗炎和抗氧化系统中十分关键。而促分裂原活化的蛋白激酶(MAPK)参与体内多种氧化应激和炎症反应过程并发挥重要调控作用。MAPK与Nrf2/HO-1信号通路间具有复杂的相互作用,且这一作用在不同的研究条件下并不一致。不同的MAPK可不同程度地影响Nrf2的活化,进而影响HO-1的表达;另外,HO-1降解胆红素的产物一氧化碳及其他Nrf2调控的基因也可能对一些MAPK具有调节作用。同时,细胞内其他信号分子的影响进一步增加了MAPK与Nrf2/HO-1信号通路相互作用的多样性。
Nuclear factor-erythroid 2-related factor 2(Nrf2)is an intracellular transcriptional regulator and Heme oxygenase-1(HO-1)is one of its most important downstream regulatory products.The cascade reaction of the two is critical in the body′s anti-inflammatory and antioxidant systems.Mitogen-activated protein kinases(MAPK)are involved in various processes of oxidative stress and inflammatory response in vivo and play an important regulatory role.There is a complex interaction between MAPK and Nrf2/HO-1 signaling pathway,which is inconsistent under different conditions of researches.Different MAPK can affect the activation of Nrf2 and thus affect the expression of HO-1.Carbon monoxide,one of the products of bilirubin degraded by HO-1 and other Nrf2 regulated genes may also have regulatory effects on some MAPK.In addition,the influence of other signaling molecules within the cell further increases the diversity of their interactions.
